Identifying the critical oncogenic mechanism of LDHA based on a prognostic model of T-cell synthetic drivers

NR3C2 inhibits the proliferation of colorectal cancer via regulating glucose metabolism and phosphorylating AMPK

Summary: We aimed to investigate the roles and mechanisms of NR3C2 in colorectal cancer (CRC). The expression of NR3C2 was significantly decreased in CRC tumors compared to paracancerous tissues, and this was correlated with distant metastasis, poor prognosis, and advanced stages of CRC patients. Overexpressing NR3C2 inhibited the proliferation of CRC cells and promoted G2/M cell cycle arrest. Furthermore, NR3C2 suppressed glucose metabolism by decreasing the expression of HK2 and LDHA, and downregulated the phosphorylation of AMPK in CRC cells. This study identified NR3C2 as a potential therapeutic target for CRC by inhibiting glucose metabolism and AMPK phosphorylation.

Summary: This study identifies APOL3 as a significant modulator for ferroptosis-related CD8+ infiltration in colorectal cancer. Through experimental validation, it is found that APOL3 binds LDHA to promote its ubiquitylation-related degradation, thereby promoting tumor ferroptosis and cytotoxic ability of CD8+ T cells. The findings provide a novel target to overcome drug resistance to ferroptosis and immunotherapy.

A genome-scale screen for synthetic drivers of T cell proliferation

Metabolic enzyme LDHA activates Rac1 GTPase as a noncanonical mechanism to promote cancer

Summary: The glycolytic enzyme lactate dehydrogenase A (LDHA) activates small GTPase Rac1 to promote cancer independently of its glycolytic enzyme activity. LDHA interacts with the active form of Rac1, Rac1-GTP, to inhibit Rac1-GTP interaction with its negative regulator, GTPase-activating proteins, leading to Rac1 activation. LDHA overexpression in clinical breast cancer specimens is associated with higher Rac1 activity. Inhibition of Rac1 suppresses the oncogenic effect of LDHA. Combination inhibition of LDHA enzyme activity and Rac1 activity displays a synergistic inhibitory effect on breast cancers with LDHA overexpression.
