Rescue of the first alphanucleorhabdovirus entirely from cloned complementary DNA: An efficient vector for systemic expression of foreign genes in maize and insect vectors

Summary: This study reports the construction of an infectious clone for the maize-infecting Alphanucleorhabdovirus maydis, which is the first efficient NSR vector for maize. The infectious clone was established using reverse genetics technologies and a GFP reporter gene was inserted to monitor virus infection. The results demonstrate the stability and efficiency of this infectious clone in plant and insect hosts.

Engineered biocontainable RNA virus vectors for non-transgenic genome editing across crop species and genotypes

Summary: CRISPR/Cas genome-editing tools have revolutionized plant biology research and crop breeding, but the lack of robust delivery methods hinders their widespread adoption in plant science. This study presents an efficient, non-transgenic CRISPR/Cas delivery platform based on the engineered tomato spotted wilt virus (TSWV), which can infect over 1000 plant species. By removing viral elements necessary for insect transmission, the TSWV-derived viral vectors enable effective and stable delivery of genome-editing tools in plants. These vectors induced efficient gene mutations and base conversions in multiple crop species, and plants with heritable mutations could be regenerated without antibiotic selection. Additionally, treating the virus-infected tissues with ribavirin during tissue culture cleared the viral vectors and increased the recovery of heritable mutations. This viral delivery system provides a promising tool for overcoming gene delivery bottlenecks in genome editing of various crop species and elite varieties.

Virtifier: a deep learning-based identifier for viral sequences from metagenomes

Summary: In this study, a deep learning-based viral identifier called Virtifier is proposed, which efficiently encodes nucleotide sequences and analyzes relationships between codons using an attention-based LSTM neural network. Experimental results show that Virtifier accurately identifies short viral sequences (<500 bp) from metagenomes and achieves comparable performance at longer lengths (>5000 bp).

Developing reverse genetics systems of northern cereal mosaic virus to reveal superinfection exclusion of two cytorhabdoviruses in barley plants

Summary: In this study, a minireplicon system of Northern cereal mosaic virus (NCMV) was developed and a recombinant virus was successfully recovered in small brown planthoppers and barley plants. Cellular superinfection exclusion was observed in barley leaves between NCMV and barley yellow striate mosaic virus (BYSMV), indicating that the two insect-transmitted cytorhabdoviruses are enemies at the cellular level during coinfections in plants.

Petabase-scale sequence alignment catalyses viral discovery

Summary: Public databases contain a vast amount of nucleic acid sequences, but efficient methods for systematic exploration of this library have been lacking. In this study, we developed a cloud computing infrastructure, Serratus, to enable ultra-high-throughput sequence alignment at the petabase scale. By searching millions of diverse samples, we identified over 10(5) novel RNA viruses and characterized their environmental reservoirs. To facilitate viral discovery, we established a comprehensive database of these data and tools. Expanding the known sequence diversity of viruses has important implications for understanding emerging pathogens and improving pathogen surveillance.

Full genome sequence of a novel iflavirus from the leafhopper Recilia dorsalis

Summary: In this study, a new virus named Recilia dorsalis iflavirus 1 (RdIV1) was discovered in a sample of R. dorsalis. Through genome sequencing and analysis, it was determined that RdIV1 belongs to the genus Iflavirus and possesses typical characteristics of iflaviruses. This classification was supported by the highest amino sequence identity to Euscelidius variegatus virus 1 (EVV-1), a member of the same genus.

Complete genome sequence of a novel nege-like virus in aphids (genus Indomegoura)
