The tumor suppressor role and ceRNA network of miR-1294 in cancer

Yunan Mao et al.

Summary: miRNAs are important regulators of gene expression, and miR-1294 has been found to be significantly down-regulated in 15 cancers and regulated by 21 upstream regulators. It affects cancer cell proliferation, migration, invasion, and apoptosis, and its target genes are involved in important signaling pathways. Low expression of miR-1294 is associated with drug resistance and poorer prognosis in several types of cancer, emphasizing its clinical significance as a tumor suppressor.

Summary: Each year, the American Cancer Society compiles data on cancer occurrence and outcomes in the United States. The latest data shows that breast and prostate cancer progress has stagnated, while lung cancer has shown improvements in survival rates. Lung cancer incidence for advanced disease has declined while localized-stage rates have increased, resulting in higher survival rates. Mortality patterns align with incidence trends, with lung cancer deaths declining rapidly, breast cancer deaths slowing, and prostate cancer deaths stabilizing.

BIRC5 regulates inflammatory tumor microenvironment-induced aggravation of penile cancer development in vitro and in vivo

Yang Zhao et al.

Summary: In this study, the overexpression of BIRC5 in penile cancer was explored, and its role in the progression of the disease was investigated. Silencing of BIRC5 inhibited the proliferation, migration, and invasion of penile cancer cells and improved tumor growth and mouse survival. Furthermore, interferon-gamma was found to aggravate penile cancer progression, while the silencing of BIRC5 reversed these effects, suggesting BIRC5 as a potential diagnostic and therapeutic target for penile cancer.

METTL3/IGF2BP3 axis inhibits tumor immune surveillance by upregulating N6-methyladenosine modification of PD-L1 mRNA in breast cancer

Weijun Wan et al.

Summary: The study found that METTL3 can post-transcriptionally upregulate PD-L1 expression in an m(6)A-IGF2BP3-dependent manner to promote stabilization of PD-L1 mRNA. This may have important implications for new and efficient therapeutic strategies in tumor immunotherapy.

Fusobacterium nucleatum reduces METTL3-mediated m6A modification and contributes to colorectal cancer metastasis

Shujie Chen et al.

Summary: The study reveals that Fusobacterium nucleatum contributes to colorectal cancer (CRC) progression by reducing global m(6)A modifications through a YAP/FOXD3/METTL3/KIF26B axis. Fusobacterium nucleatum is closely associated with the development of CRC.

The Role of Cystine/Glutamate Antiporter SLC7A11/xCT in the Pathophysiology of Cancer

Nidhi Jyotsana et al.

Summary: SLC7A11 is an antiporter that mediates the exchange of extracellular cystine for intracellular glutamate. It plays diverse functional roles in maintaining redox homeostasis and immune system function, and is associated with the development and drug resistance of diseases such as cancer. In this review, the focus is on discussing its molecular functions and therapeutic strategies in gastrointestinal cancers.

METTL3 Inhibits Antitumor Immunity by Targeting m6A-BHLHE41-CXCL1/CXCR2 Axis to Promote Colorectal Cancer

Huarong Chen et al.

Summary: This study identifies METTL3 as a potential therapeutic target for CRC immunotherapy. Silencing or inhibiting METTL3 reduces MDSC accumulation, sustains activation and proliferation of CD4(+) and CD8(+) T cells, and ultimately suppresses CRC development. Inhibition of METTL3 plus anti-PD1 treatment shows promising antitumor efficacy against CRC.

Inhibition of USP1 enhances anticancer drugs-induced cancer cell death through downregulation of survivin and miR-216a-5p-mediated upregulation of DR5

Seon Min Woo et al.

Summary: This study found that USP1 is highly expressed in tumor tissues and is associated with poor prognosis in RCC. Mechanistically, USP1 enhances survivin stabilization by removing ubiquitin and upregulates DR5 expression by decreasing miR-216a-5p expression at the post-transcriptional level. Inhibition of USP1 sensitizes cancer cells and reduces tumor size.

METTL3 acetylation impedes cancer metastasis via fine-tuning its nuclear and cytosolic functions

Yuanpei Li et al.

Summary: In this study, the authors identify an acetylation-mediated regulation of METTL3 subcellular localization and its impact on breast cancer metastasis. They also uncover an IL-6-dependent positive feedback axis that facilitates nuclear METTL3 functions, promoting metastasis. Additionally, the study shows that aspirin treatment and SIRT1 inhibition can counterbalance the acetylation-dependent regulation of METTL3 localization and impair m(6)A methylation, leading to reduced lung metastasis.
