ICAM-1-mediated adhesion is a prerequisite for exosome-induced T cell suppression

Wei Zhang et al.

Summary: This study reveals the co-localization of ICAM-1 and PD-L1 on exosomes, both of which are upregulated by interferon-y. The adhesion between TEVs and T cells mediated by ICAM-1-LFA-1 is crucial for exosomal PD-L1-mediated immune suppression.

Noninvasive biomarkers for the diagnosis and management of autoimmune hepatitis

Claire Harrington et al.

Summary: Autoimmune hepatitis is a rare disease with unclear etiology and unpredictable response to treatment. Current treatment regimens have adverse effects and reduce quality of life. Therefore, finding biomarkers that can predict disease progression, identify relapse risk, and improve monitoring and medication dosing is of great clinical importance.

Mesenchymal stromal cells: Putative microenvironmental modulators become cell therapy

Summary: MSCs have shown exceptional safety in clinical trials, but reliable potency assays to predict their immunosuppressive efficacy are still lacking. They are approved for certain diseases in Japan and Europe, but not in the United States. Researchers discuss potential mechanisms of action, experimental models, and approaches for identifying MSC effects in vivo.

Eradication of specific donor-dependent variations of mesenchymal stem cells in immunomodulation to enhance therapeutic values

Chunxue Zhang et al.

Summary: This study revealed variations in cell proliferation and immune modulatory functions of MSCs from different donors, with IFN-gamma and NF-kappa B signaling being positively associated with immune modulatory function. Activation of these pathways could eradicate donor-dependent variations, but overall transcriptomic differences between donor-derived MSCs still exist.

One repeated transplantation of allogeneic umbilical cord mesenchymal stromal cells in type 1 diabetes: an open parallel controlled clinical study

Jing Lu et al.

Summary: The study demonstrated that one repeated intravenous dose of allogeneic UC-MSCs is safe in people with recent-onset T1D and may result in better islet beta cell preservation during the first year after diagnosis compared to standard treatment alone. The MSC-treated group showed a significantly higher rate of achieving clinical remission compared to the control arm, with some individuals even achieving insulin independence. Multivariable logistic regression analysis showed that lower fasting C-peptide and higher dose of UC-MSC correlated with the achievement of clinical remission after transplantation, with no severe side effects observed.
