c-Abl Tyrosine Kinase Is Required for BDNF-Induced Dendritic Branching and Growth

Summary: Brain-derived neurotrophic factor (BDNF) activates TrkB receptor and downstream pathways, promoting neuronal survival, growth, and plasticity. The role of c-Abl, a non-receptor tyrosine kinase, in BDNF-induced dendritic arborization was examined using various strategies. Surprisingly, BDNF promoted c-Abl activation and interaction with TrkB receptors. Inhibition or genetic ablation of c-Abl abolished BDNF-induced dendritic arborization without affecting classic TrkB downstream pathways.

Neurophysiology of the Developing Cerebral Cortex: What We Have Learned and What We Need to Know

Summary: This review article provides a brief summary of the novel technologies, challenges, current understanding, and open questions in the field of neurophysiology of the developing cerebral cortex in rodents. The use of high-density multi-electrode arrays and genetically encoded calcium indicators has allowed the recording of neuronal activity in large cortical networks, revealing surprising patterns of cortical and subcortical activity during early development. However, the functional roles of these activity patterns and their impact on cortical maturation are not yet fully understood. Future research will focus on targeted genetic manipulations and optogenetics to elucidate the function of early cortical circuits and their role in the generation of activity patterns.

Mutant CRMP1-Tyr504Phe suppresses Sema3A-induced growth cone collapse and dendritic growth in mouse neurons.

Extrinsic Factors Regulating Dendritic Patterning

Summary: Stereotypic dendrite arborizations play a crucial role in neuronal identity, determining synaptic input fields and information integration within neural circuits. Extrinsic factors, including adhesion receptors and secreted proteins from surrounding neurons or supporting cells, regulate dendrite patterning through local cytoskeletal and adhesion modulation as well as global transcriptional regulation. The coordinated signaling of multiple extrinsic factors promotes complexity in dendritic trees and ensures robust dendritic patterning.

Summary: Reelin plays a crucial role in postnatal interneuron development by regulating dendritic growth, with deficiency leading to hypertrophy. Dysfunction of presynaptic GABA(B)Rs in interneurons lacking Reelin may contribute to the dendritic hypertrophy. These findings highlight the importance of Reelin in signaling to halt excessive growth during postnatal interneuron maturation.

Summary: Mutations in the GRIN2B gene lead to autism spectrum disorders, with a GluN2B variant affecting dendrite morphogenesis by shifting branch growth towards retraction. Mutant neurons exhibit increased pruning of dendritic branches despite forming new branches at similar rates to wild-type neurons. This results in a nearly complete elimination of the net expansion of arbor size and complexity during developmental periods.
