A pluripotent developmental state confers a low fidelity of chromosome segregation

Summary: During in vitro propagation, human pluripotent stem cells (hPSCs) frequently become aneuploid with improper chromosome numbers due to mitotic errors. The primary cause of these errors is lagging chromosomes in anaphase with improper merotelic microtubule attachments. Short-term treatment (<24 h) with small molecules that affect mitotic duration or chromosome microtubule attachments can reduce these errors, although hPSCs adapt and the errors rebound upon long-term (>24 h) treatment. Mitotic error rates correlate with developmental potential, increasing or decreasing upon loss or gain of pluripotency, respectively. Thus, hPSCs exhibit a low mitotic fidelity, and chromosome segregation fidelity is dependent on the developmental state in normal human cells.

Cultivated autologous limbal epithelial cell (CALEC) transplantation: Development of manufacturing process and clinical evaluation of feasibility and safety

Summary: In this study, a novel xenobiotic-free, serum-free, antibiotic-free, two-step process was developed to treat unilateral limbal stem cell (LSC) deficiency. Cultivated autologous limbal epithelial cells (CALEC) were generated from limbal biopsies and expanded onto human amniotic membrane with rigorous quality control. The clinical trial showed preliminary feasibility and safety of CALEC transplants in patients with unilateral LSC deficiency, leading to a second phase of recruitment to collect longer term safety and efficacy data.
