Variation in CD8 T cell IFNγ differentiation to strains of Toxoplasma gondii is characterized by small effect QTLs with contribution from ROP16

Summary: Toxoplasma gondii induces a strong CD8 T cell response characterized by the secretion of IFN gamma. The initiation of CD8 T cell IFN gamma responses differs between clonal lineage strains, with type I strains being low inducers and types II and III strains being high inducers. Our study suggests that CD8 T cell IFN gamma production to T. gondii strains is regulated by multiple polymorphisms, including ROP16, which can influence the commitment of responding CD8 T cells to IFN gamma production.

Proximity-Labeling Reveals Novel Host and Parasite Proteins at the Toxoplasma Parasitophorous Vacuole Membrane

Summary: In this study, the researchers employed human foreskin fibroblasts expressing a proximity-labeling enzyme and quantitative proteomics to identify novel parasite and host proteins localized at the Toxoplasma parasitophorous vacuole membrane (PVM) interface. The results revealed the presence of three parasite proteins and four host proteins at the PVM in infected human cells, expanding our understanding of the molecular players at this crucial host-pathogen interface. These findings suggest the presence of novel biology at the Toxoplasma PVM, particularly with the identification of host proteins involved in the ESCRT machinery.

Toxoplasma-proximal and distal control by GBPs in human macrophages

Summary: Human guanylate binding proteins (GBPs) are important for cell intrinsic defense mechanisms against intracellular pathogens. GBP1, GBP2, and GBP5 play different roles in controlling Toxoplasma gondii infection, with GBP1 acting directly on the parasitophorous vacuole and GBP2/5 affecting the growth of the parasite from a distance. Mutations in GBP1/2/5 can lead to mis-localization and impair their function in parasite control.

Summary: This study used the Oxford Nanopore Minion platform to generate near-complete de novo genome assemblies for multiple strains of Toxoplasma gondii and Neospora caninum, significantly improving genome contiguity and resolving hundreds of tandem repeats. Although both species retained a 13-chromosome karyotype, extensive chromosome-scale rearrangements were found to have occurred in both Toxoplasma gondii and Neospora caninum since their most recent common ancestry.
