Molecular diagnostic tools for the World Health Organization (WHO) 2021 classification of gliomas, glioneuronal and neuronal tumors; an EANO guideline

Felix Sahm et al.

Summary: In the 5th edition of the WHO CNS tumor classification, molecular characteristics have become important diagnostic criteria for many CNS tumor types. This guideline focuses on the methods used for diagnosing gliomas, glioneuronal, and neuronal tumors using informative molecular markers. Various molecular methods are discussed, including next-generation sequencing, methylation profiling, and immunohistochemistry. The guideline also covers the analysis of MGMT promoter methylation status, and provides an overview of the advantages, limitations, and requirements of different assays. The importance of molecular diagnostic testing in neuro-oncology is emphasized, along with discussions on clinical relevance, accessibility, cost, and ethical aspects.

Oncogenic structural aberration landscape in gastric cancer genomes

Mihoko Saito-Adachi et al.

Summary: This study analyzes the structural rearrangement landscape of 170 gastric cancer genomes and identifies six rearrangement signatures. Specific rearrangement signatures are associated with driver events and epidemiological backgrounds. Additionally, 27 structural variant hotspots potentially linked to gastric cancer driver genes are discovered.

Bulk and single-cell RNA-sequencing analyses along with abundant machine learning methods identify a novel monocyte signature in SKCM

Yuyao Liu et al.

Summary: In this study, a novel eight-gene monocyte-related signature (MRS) was identified as a prognostic biomarker for skin cutaneous melanoma (SKCM) patients. The MRS showed better accuracy in predicting patient outcomes compared to traditional clinical variables and molecular features. The IFITM3 gene was also identified as a potential biomarker for SKCM.

Endometrial Expression of Cell Adhesion Genes in Recurrent Implantation Failure Patients in Ongoing IVF Cycle

Amruta D. S. Pathare et al.

Summary: Recurrent implantation failure (RIF) is a major obstacle in IVF treatment. A study was conducted to analyze gene expression in non-pregnant RIF patients vs fertile women under controlled ovarian stimulation (COS), revealing significant downregulation of cell adhesion function and confirming downregulation of six out of eight related genes in RIF patients. These findings suggest the importance of these genes in the endometrial receptivity (ER) mechanism under COS. Additional analysis in larger cohorts is needed to validate the study findings in RIF patients undergoing IVF.

The Clinical Efficacy of Personalized Embryo Transfer Guided by the Endometrial Receptivity Array/Analysis on IVF/ICSI Outcomes: A Systematic Review and Meta-Analysis

Zhenteng Liu et al.

Summary: The study evaluated the prevalence of WOI displacement in infertile women and the impact of ERA testing on IVF/ICSI outcomes. It found that approximately one third of infertile women could have WOI displacement, and ERA testing significantly increases the chances of successful pregnancy in non-receptive patients with RIF.

Single-cell analysis of menstrual endometrial tissues defines phenotypes associated with endometriosis

Andrew J. Shih et al.

Summary: This study found significant differences in endometrial tissues between endometriosis cases and controls. The controls had more proliferating uterine natural killer cells and decidualized endometrial stromal cells, while the cases had more cells expressing inflammatory and senescent phenotypes.

A novel platform for discovery of differentially expressed microRNAs in patients with repeated implantation failure

Ching Hung Chen et al.

Summary: The study identified predictor microRNAs in patients with repeated implantation failure (RIF) by analyzing miRNA profiles from the endometrium of patients undergoing in vitro fertilization (IVF). A signature set of miRNAs associated with the risk of RIF was identified, and a 3-miRNA signature was able to predict patients with RIF with an accuracy >90% in the validation cohort. The findings suggest that specific endometrial miRNAs can be used as diagnostic biomarkers to predict RIF and potentially increase the success rate of implantation practice.

Single-Cell Transcriptomics: Current Methods and Challenges in Data Acquisition and Analysis

Asif Adil et al.

Summary: Rapid advancements in next-generation sequencing have made single-cell transcriptomics a conventional practice in scientific laboratories, providing new insights into the heterogeneity of cells at the individual level. Challenges in analyzing the data include normalization, differential gene expression analysis, and dimensionality reduction. New methods like 10x Chromium can profile millions of cells in parallel, creating a considerable amount of data and highlighting the need for new bioinformatics algorithms and tools for analysis.

Endometrial Factor in Unexplained Infertility and Recurrent Implantation Failure

Xavier Santamaria et al.

Summary: Unexplained infertility and recurrent implantation failure are diagnoses made based on failed pregnancy attempts, often leading to frustration for patients and caregivers. Evidence suggests that endometrial factors may play a role in these cases, potentially improving treatment outcomes.
