Transport engineering in microbial cell factories producing plant-specialized metabolites

Zeinu Mussa Belew et al.

Summary: Transport engineering strategies alter the expression of transporter proteins to change the distribution of metabolites within an organism. The production of plant specialized metabolites in microbial cell factories faces challenges such as premature pathway termination, feedback inhibition, low yields, and poor connectivity between subcellular compartments. This article highlights recent examples of transport engineering in microbial cell factories and proposes future research directions to advance the field.

Growth Inhibition by Amino Acids in Saccharomyces cerevisiae

Stephanie J. Ruiz et al.

Summary: Amino acids are essential for metabolism but can be toxic at high levels intracellularly. Downregulation of amino acid transporters in Saccharomyces cerevisiae is a mechanism to avoid toxicity. Overexpression of certain amino acid transporters can induce growth defects, and various amino acids are growth inhibitory to yeast cells when transported into the cell at high levels.

Transportome-wide engineering of Saccharomyces cerevisiae

Guokun Wang et al.

Summary: Synthetic biology enables the production of small molecules by recombinant microbes for various applications. A method for genome-wide transporter disruption combined with a metabolite biosensor was described to identify transporters impacting the production of specific metabolites in yeast. This approach facilitates high-throughput transporter identification for small molecules in cell factories.

Yeast Double Transporter Gene Deletion Library for Identification of Xenobiotic Carriers in Low or High Throughput

Ludimila Dias Almeida et al.

Summary: By utilizing a library of double transporter deletion strains, potential drug import and export routes can be rapidly identified, aiding in determining the necessary chemical groups for transport via specific carriers. This information can lead to the development of drugs with optimal absorption and reduced likelihood of resistance.

The Transporter Classification Database (TCDB): 2021 update

Milton H. Saier et al.

Summary: The Transporter Classification Database (TCDB) is a freely accessible reference resource providing comprehensive information about transporters from various organisms, and is the only transport protein classification database adopted by the International Union of Biochemistry and Molecular Biology. The database has seen a significant increase in proteins and transport systems since its last update in 2016, with new features including a chemical ontology for substrates, new superfamilies, and novel subclasses of transport systems.
