Drug repurposing for cardiovascular diseases: New targets and indications for probenecid

Zsofia Onodi et al.

Summary: Pharmacological options for cardiovascular diseases have been effective in slowing disease progression, but long-term prognosis remains poor, necessitating new therapeutic strategies. Drug repurposing offers approved and abandoned drugs new indications, including gout-related inflammation drugs like canakinumab, colchicine, and uricosuric drug probenecid. Probenecid has been found to modulate myocardial contractility, vascular tone, and exert anti-inflammatory effects. This review summarizes experimental and clinical data on repurposing probenecid for ischaemic heart disease and subsequent heart failure and suggests its potential for treating cardiovascular diseases.

Summary: This study provides evidence that I2R does not play a functional role in modulating vascular tone in mouse aorta. MCR5, an I2R ligand, acts as an endothelium-independent vasodilator and is resistant to aging.

Summary: The TRP ion channel family consists of 27 channel proteins, with TRPV subfamily being a recent focus in cardiovascular research, particularly TRPV1 and TRPV2. Studies on TRPV2 have shown its significance in cardiovascular function, with clinical research indicating its role in patients with muscular dystrophy, cardiomyopathy, and heart failure with reduced ejection fraction.
