JAK2 inhibitor ameliorates the progression of experimental autoimmune myasthenia gravis and balances Th17/Treg cells via regulating the JAK2/STAT3-AKT/mTOR signaling pathway

Yaru Lu et al.

Summary: This study demonstrates that JAK2 inhibitors can ameliorate myasthenia gravis (MG) by regulating the Th17/Treg balance. The results show that JAK2 inhibitors reduce immune responses in EAMG rats, improve clinical symptoms, and increase AChR aggregation in the postsynaptic membrane. Additionally, the study reveals crosstalk between the JAK2/STAT3 and AKT/mTOR pathways in EAMG rats.

Regulatory T Cells and Inflammatory Mediators in Autoimmune Disease

Victoire Gouirand et al.

Summary: Regulatory T cells play a critical role in regulating tissue inflammation, and loss of their suppressive capacity may contribute to autoimmune disease. Under specific conditions, these cells can lose their ability to suppress and release proinflammatory cytokines, promoting inflammation. The impact of cytokines on Treg function may have implications for autoimmune disease.

HIF-1α aggravated traumatic brain injury by NLRP3 inflammasome-mediated pyroptosis and activation of microglia

Dong Yuan et al.

Summary: HIF-1α is involved in regulating neuronal death after TBI and inflammation, as shown in a study exploring the regulatory mechanism between HIF-1α and NLRP3 inflammasome. TBI-induced brain injury is characterized by edema, blood-brain barrier disorder, and an upregulation of HIF-1α expression. Inactivation of HIF-1α provided neuroprotection and alleviated brain injury post-TBI by attenuating NLRP3 inflammasome-mediated pyroptosis.

Gut Microbiota as Regulators of Th17/Treg Balance in Patients With Myasthenia Gravis

Pan Chen et al.

Summary: Myasthenia gravis is an acquired neurological autoimmune disorder characterized by dysfunctional transmission at the neuromuscular junction, with immune imbalance playing a role in its pathogenesis. Gut microbiota has been identified as a significant factor influencing the balance of Th17/Treg cells in MG patients.

Summary: Germinal center reactions are crucial for the correct functioning of the adaptive immune system, leading to the formation of high affinity, class switched antibodies. Through a dynamic process involving various cells and molecules, B cells undergo differentiation into memory B cells and plasma cells, ultimately enhancing the immune response.
