A labile iridium(III) complex-based luminogenic probe for the ratiometric detection of dithiocarbamate compounds in living system

Summary: In this study, an iridium(III) solvento complex (1) was developed for the luminogenic detection of dithiocarbamates. The probe showed high sensitivity and selectivity towards various dithiocarbamates including NDDC, SDDC, and APDC. It could detect dithiocarbamates in aqueous buffer, water, drink samples, and even in the presence of living cells.

A novel pyrimidine-based two-photon fluorogenic probe for rapidly visualizing nitroreductase activity in hypoxic cancer cells and in vivo

Summary: This study developed a NTR probe (ZJ) that can accurately and rapidly detect the overexpressed NTR activity in tumor cells. ZJ showed responsive turn-on two-photon excited fluorescence by NTR recognition in both cells and in vivo. The probe demonstrated favorable stability and specificity, and successfully visualized the distribution of overexpressed NTR in hypoxic cancer cells and zebrafish, indicating its potential application in studying the relationship between tumor hypoxic microenvironment and cancer.

Phosphorogenic Iridium(III) bis-Tetrazine Complexes for Bioorthogonal Peptide Stapling, Bioimaging, Photocytotoxic Applications, and the Construction of Nanosized Hydrogels

Summary: 1,2,4,5-tetrazine functions as both a bioorthogonal reactive unit and a luminescence quencher, making it a desirable candidate for luminogenic labeling in living systems. This study synthesized and characterized three cyclometalated iridium(III) complexes with two tetrazine units, which exhibited high reaction kinetics towards specific substrates, allowing effective peptide labeling. These complexes were also applied for live cell bioimaging and photocytotoxicity, and one of them was used to prepare luminescent nanosized hydrogels with interesting cargo delivery properties.

Recent advances in the fluorescent probes for flavinase activity: design and applications

Summary: Flavinases, including various redox enzymes, play important roles in organisms and their aberrant expression is related to various diseases. Accurate measurement of flavinases is crucial for the early diagnosis of diseases. Fluorescence probes, known for their easy and accurate sensing capability, have drawn great attention for detecting flavinases. This review introduces the recent advances in the design and application of flavinase probes, analyzes the design strategies and reaction mechanisms, and discusses the current challenges, which will further advance the development of diagnostic and therapeutic approaches for flavinase-related diseases.

Phosphorescent iridium(III) complexes as lifetime-based biological sensors for photoluminescence lifetime imaging microscopy

Summary: Luminescent iridium(III) complexes have been extensively studied as intracellular sensors for photoluminescence microscopy imaging due to their high phosphorescence quantum yields, photostability, good cell membrane permeability, and rich and sensitive photophysical properties. Photoluminescence lifetime imaging microscopy (PLIM) allows for visualizing and mapping the decay of luminescent dyes in the time domain during cellular imaging. This review article focuses on the design and applications of lifetime-responsive phosphorescent iridium(III) complexes as imaging reagents for mapping environmental parameters and sensing analytes of interest in intracellular conditions.

MicroPET imaging of bacterial infection with nitroreductase-specific responsive 18F-labelled nitrogen mustard analogues

Summary: This study successfully developed two novel radiotracers, F-18-NTRP and F-18-NCRP, for imaging deep-seated bacterial infections using positron emission tomography (PET). The tracers showed high selectivity and specificity for nitroreductase (NTR) response and effectively distinguished infection from sterile inflammation in animal models. Further investigation is needed to explore their potential clinical application.
