BST2 induced macrophage M2 polarization to promote the progression of colorectal cancer

Single-cell landscape and clinical outcomes of infiltrating B cells in colorectal cancer

Summary: In this study, the characteristics and clinical significance of B cells in colorectal cancer (CRC) were investigated using single-cell RNA sequencing and multicolour immunofluorescence staining experiments. Five distinct subtypes of B cells were identified in the CRC tumour microenvironment, along with their marker genes, distribution patterns and functional properties. IgG plasma cells were found to be more abundant in tumour sites compared to adjacent normal tissues. Additionally, it was discovered that CXCL13-producing CD8(+) T cells and the CCL28-CCR10 axis play important roles in the formation and migration of B cells in the tumour microenvironment. The study also identified four distinct colon immune classes, with CD20(+) B cells being enriched in an immune-inflamed tumour group. This B cell-rich group demonstrated strong antigen presentation, accumulation of IgG plasma cells, high microsatellite instability and tumour mutation burden, as well as immunosuppressive properties, making it a potential predictive biomarker for future immunotherapy. Furthermore, patients with enrichment of B cells and tertiary lymphoid structures (TLSs) were shown to have significant therapeutic advantages in an immunotherapy cohort. Overall, these findings provide a detailed landscape of infiltrating B cells in CRC and their potential clinical significance.

Quiescent cancer cells resist T cell attack by forming an immunosuppressive niche

Summary: This study discovered that in primary breast cancer, tumor cells that resist T cell attack are quiescent and form clusters with reduced immune infiltration. These quiescent cancer cells show superior tumorigenic capacity and higher expression of chemotherapy resistance and stemness genes. Single-cell RNA sequencing revealed differential phenotypes in infiltrating cells based on their intra-tumor location, and uncovered hypoxia-induced immune-suppressive programs. The findings suggest that eliminating quiescent cancer cells holds the promise to counteract immunotherapy resistance and prevent disease recurrence in TNBC.
