Identification of HPCAL1 as a specific autophagy receptor involved in ferroptosis

Xin Chen et al.

Summary: This study identifies HPCAL1 as a novel autophagy receptor for the selective degradation of CDH2 during ferroptosis. Depletion of CDH2 increases susceptibility to ferroptotic death. The phosphorylation of HPCAL1 and the non-classical LC3-interacting region motif play key roles in the autophagic degradation of CDH2. A ferroptosis inhibitor is found to suppress HPCAL1 expression. Inhibition of HPCAL1 prevents ferroptosis-induced tumor suppression and pancreatitis in mouse models.

Genistin modulates high-mobility group box protein 1 (HMGB1) and nuclear factor kappa-B (NF-κB) in Ehrlich-ascites-carcinoma-bearing mice

Mohamed A. Saleh et al.

Summary: Cancer is the second leading cause of death worldwide, and current treatment options are often uncomfortable and ineffective. Plant-derived natural chemicals, such as flavonoids, have gained attention due to their high bioavailability, safety, few side effects, and cost-effectiveness. Flavonoids have shown promising cytotoxic anti-cancer effects and are being researched for their ability to target inflammation and apoptosis. In a laboratory study, genistin, a flavonoid compound, demonstrated good cytotoxic activity against breast cancer cell lines. In vivo studies on animals also showed that genistin reduced tumor weight, volume, and certain biochemical markers associated with cancer progression.

HMGB1 is a mediator of cuproptosis-related sterile inflammation

Summary: This study found that cuproptotic cells release HMGB1 to initiate inflammation. Mechanistically, copper accumulation-induced ATP depletion activates AMPK to promote HMGB1 phosphorylation, resulting in increased release. Inhibiting AMPK activation reduces cuproptosis and HMGB1 release. Cuproptotic cells deficient in HMGB1 show reduced production of inflammatory cytokines. Thus, HMGB1 is a key mediator of sterile inflammation initiated by cuproptosis.

Inhibition of the Receptor for Advanced Glycation End Products Enhances the Cytotoxic Effect of Gemcitabine in Murine Pancreatic Tumors

Priyanka Swami et al.

Summary: Studies have shown that RAGE plays a critical role in promoting pancreatic cancer cell proliferation and resistance to gemcitabine. Blocking the RAGE/HMGB1 interaction can enhance the cytotoxic effect of gemcitabine, reduce autophagy, and protect against excessive weight loss during treatment.

Mechanism of interaction between autophagy and apoptosis in cancer

Shreya Das et al.

Summary: Autophagy and apoptosis play significant roles in cancer, but they may exhibit dual effects on cancer cell survival as the disease progresses. Interactions between a complex array of biomarkers are crucial in maintaining coordination between the two pathways, influencing the survival or death of malignant cells.

Calcium Signaling Regulates Autophagy and Apoptosis

Pramod Sukumaran et al.

Summary: Calcium serves as a crucial second messenger in regulating various physiological functions, such as cell growth/development, cell survival, and programmed cell death processes. The homeostasis of cytosolic calcium levels, maintained through coordination of proteins/pumps/Ca2+ channels and Ca2+ storage in organelles, is essential for cellular functions. Store operated Ca2+ entry (SOCE) mechanism, activated by calcium depletion, influences functions in both excitable and non-excitable cells, with potential implications in neurodegenerative conditions.
