T-Cell Receptor Repertoire Sequencing in the Era of Cancer Immunotherapy

Meredith L. Frank et al.

Summary: T cells, with unique T-cell receptors (TCR) that recognize specific antigens, play a critical role in the adaptive immune system. Analyzing the T-cell repertoire using next-generation sequencing technologies provides insights into immune responses and diseases like cancer. This review article provides an overview of TCR sequencing, covering library preparation, sequencing and analysis methods, and functional validation techniques. It also discusses important TCR repertoire metrics, computational tools for predicting antigen specificity, and the application of TCR sequencing in understanding tumor biology and developing cancer immunotherapies.

Complement System: An Immunotherapy Target in Colorectal Cancer

Iman M. Talaat et al.

Summary: Colorectal cancer is the third most common malignant tumor globally, and the complement system of the immune system plays a significant role in combating cancer. This review discusses complement system activation in CRC and its interactions with TME components, providing a framework for further understanding the potential therapies targeting complement activation in solid tumors such as CRC.

Survival impact of immune cells infiltrating peritumoral area of hepatocellular carcinoma

Toshihiko Yusa et al.

Summary: Inflammatory and immune cells in the tumor microenvironment are associated with tumor progression and prognosis in hepatocellular carcinoma (HCC) patients. The expression of tumor-associated neutrophils (TANs), M2 macrophages (TAMs), CD8(+) T cells, and regulatory T cells (Tregs) in HCC patients was evaluated, and their relationships with clinicopathological characteristics and survival outcomes were analyzed.

Revised diagnostic criteria for neurofibromatosis type 1 and Legius syndrome: an international consensus recommendation

Eric Legius et al.

Summary: The study revised the diagnostic criteria for neurofibromatosis type 1 (NF1) and established criteria for Legius syndrome (LGSS) by combining major developments in genetics, ophthalmology, dermatology, and neuroimaging. Consensus on minimal clinical and genetic criteria for diagnosing and differentiating NF1 and LGSS was reached, with recommendations for mosaic forms of these conditions. Continued refinement of these new criteria will be necessary to study their diagnostic properties, reconsider criteria not included, and identify new features of the conditions, hence proposing an initiative to periodically update the diagnostic criteria for NF1 and LGSS.
