Immunomodulatory role of azithromycin: Potential applications to radiation-induced lung injury

Summary: Radiation-induced lung injury (RILI), a side effect of radiotherapy for thoracic tumors, can result in pneumonitis and pulmonary fibrosis. Azithromycin, with its immunomodulatory and anti-inflammatory effects, has shown potential benefits for RILI patients in preclinical studies. Clinical studies have also suggested that azithromycin may benefit patients with acute lung injury due to its anti-inflammatory and immunomodulatory effects.

CC Chemokines in Idiopathic Pulmonary Fibrosis: Pathogenic Role and Therapeutic Potential

Summary: Idiopathic pulmonary fibrosis (IPF) is a chronic and progressive respiratory disease characterized by irreversible decline in lung function. The dysregulated expression of chemokines and chemokine receptors has been associated with the development and progression of IPF. Chemokines of the CC family play significant roles in exacerbating IPF progression. Modulating levels of detrimental CC chemokines and interrupting the corresponding transduction axis by neutralizing antibodies or antagonists are potential treatment options for IPF. This review focuses on the roles of different CC chemokines in IPF pathogenesis and their potential use as biomarkers or therapeutic targets.

MiR-223-3p attenuates radiation-induced inflammatory response and inhibits the activation of NLRP3 inflammasome in macrophages

Summary: This study investigated the mechanisms of NLRP3-mediated macrophage pyroptosis and identified the role of miR-223-3p in regulating NLRP3 expression. It was found that miR-223-3p can decrease the expression of NLRP3 and proinflammatory factors, resulting in reduced pyroptosis in irradiated murine macrophages. In vivo experiments further demonstrated the effectiveness of miR-223-3p supplementation in alleviating acute radiation-induced lung injury.

The occurrence and development of radiation-induced lung injury after interstitial brachytherapy and stereotactic radiotherapy in SD rats

Summary: This study compared the severity of radiation-induced lung injury (RILI) after interstitial brachytherapy and stereotactic radiotherapy (SBRT). The results showed that the CT values of the right lung in the SBRT group were significantly higher than those in the control group and the interstitial brachytherapy group. The expressions of IFN-γ, IL-2, IL-6, and IL-10 in the SBRT group were significantly higher than those in the interstitial brachytherapy group. The TGF-β expression in the interstitial brachytherapy group reached its peak and was significantly lower than the SBRT group. The mortality rate in the SBRT group was significantly higher than that in the interstitial brachytherapy group.

The pathophysiology of fibrosis involves several factors such as TGF-B, vascular epithelial growth factor, fibroblast growth factor, and platelet-derived growth factor. Type-17 immunity, activated by cytokines like IL-1B, IL-6, and IL-23, plays a role in pulmonary fibrosis. Targeting type-17 immunity may be a potential therapeutic strategy for preventing or exacerbating pulmonary fibrosis.
