Theoretical Aspects and Applications of Aqueous Two-Phase Systems

Summary: The aqueous two-phase system (ATPS) is a technique that produces high-purity products in a single step and is particularly useful for purifying biomolecules. Aqueous two-phase extraction (ATPE) involves transferring solutes between two aqueous phases, typically using polymer-polymer or polymer-salt systems. This review covers recent developments in ATPS for biomolecule recovery, including the mechanism of phase formation, conditions for solute partitioning, and factors influencing ATPS, such as polymer concentration, salt type, pH, and temperature. The increasing applications of ATPS for high-value bioproduct recovery and the development of low-cost alternatives are also discussed.

Simultaneous extraction and enrichment of sunset yellow dye in an aqueous two-phase system

Summary: This study analyzes the extraction of Sunset Yellow (SSY) dye using a polymer-salt aqueous two-phase system (ATPS) as a green alternative to organic solvent-based extraction processes. The results show that by using polyethylene glycol and sodium sulfate as the polymer and salt phases, respectively, the dye can be selectively transferred and extracted. Experimental results demonstrate that 75% extraction can be achieved in 2 hours under diffusion-controlled conditions, while using a milli channel can accelerate the process and achieve 77% extraction in around 3 minutes. The use of slug flow in the microchannel helps improve mass transfer, leading to 85% extraction in the PEG6000-salt system in 3 minutes.

Extraction and preconcentration of vanadium from food samples using aqueous two-phase systems by a multivariate study

Summary: This study proposes a new method for the determination of vanadium in food samples using an aqueous two-phase system. By applying multivariate analysis and Box-Behnken matrix, the significant variables and critical points for extraction were defined. The developed methodology showed accurate and efficient results, with detection limits of 0.0220 μg/kg and quantification limits of 0.665 μg/kg.

Equilibrium phase behavior of ternary systems formed by polyethylene glycol plus choline salts plus water

Summary: The economic and environmental potential of choline salts in aqueous two-phase systems (ATPS) combined with polyethylene glycol (PEG) has been studied. New green ATPS formed by PEG and choline-based ionic liquids (ILs) were characterized. It was found that the behavior of choline chloride is different from bitartrate and dihydrogen citrate. ATPS formed by PEG1500 showed a significant increase in the biphasic area with temperature, while PEG400 showed a slight increase, indicating an endothermic phase segregation. The binodal curves and tie-lines were successfully fitted using the Merchuk and Guan equations, respectively.

Short-chain alcohol/salt-based aqueous two-phase system as a novel solvent for extraction of plant active ingredients: A review

Summary: This review summarizes the research progress of short-chain alcohol/salt-based aqueous two-phase systems (ATPS) for the recovery of plant active compounds. It includes the principle and mechanism, the main factors affecting the efficiency, recent applications, and future perspectives. The findings show that alcohol/salt-based ATPS can significantly improve the purity of extracts and achieve higher extraction efficiency with the aid of ultrasound or microwave. Therefore, alcohol/salt-based ATPS plays a crucial role in improving the recovery of plant active ingredients and has broader application prospects in industrial production.

Thermodynamic Studies of L-Tryptophan and L-Threonine Partitioning in Aqueous Two-phase Systems Containing Deep Eutectic Solvents (Choline Chloride/PEG) and Potassium Salts

Summary: Deep eutectic solvents (DES) are a safe alternative for separating biological materials like amino acids. This study investigated the phase diagrams of DES mixed with potassium salts, as well as the extraction efficiency of L-tryptophan and L-threonine at 298.15 K. The interaction between amino acids and DES was justified using volumetric results obtained from ternary and binary systems.

Liquid-liquid equilibrium measurements and computational study of salt-polymer aqueous two phase system for extraction of analgesic drugs

Summary: In this study, an aqueous two phase system was prepared using polyethylene glycol and potassium hydroxide, and its phase diagram and extraction performance were investigated. The results showed that the extraction efficiency of drugs was related to their hydrophobicity and partition coefficient. The interaction between drugs and polyethylene glycol was also studied, and the results were consistent with the extraction efficiency trend.
