Uniform thin ice on ultraflat graphene for high-resolution cryo-EM

Summary: Cryo-electron microscopy (cryo-EM) is used to visualize the atomic structure of macromolecules in their native state by embedding them in thin ice. However, controlling the uniformity of ice thickness is a challenge in high-resolution cryo-EM. In this study, we found that using ultraflat graphene (UFG) as the supporting film improves control over vitreous ice thickness. We successfully determined the 3D structures of hemoglobin, alpha-fetoprotein, and streptavidin using this method.

Summary: The ATP-binding cassette transporter ABCA3 plays a critical role in pulmonary surfactant synthesis. Mutations in human ABCA3 are the most common cause of inherited surfactant dysfunction disorders. This study presents the cryo-EM structures of human ABCA3 in two conformations and reveals a potential substrate binding mechanism and a conserved lateral access and extrusion mechanism for lipid transport mediated by ABCA transporters.

α-Fetoprotein fragment synergizes with sorafenib to inhibit hepatoma cell growth and migration and promote the apoptosis

Summary: This study presents a new strategy for treating hepatocellular carcinoma (HCC) by inhibiting its malignant behavior with AFP fragments. A specific AFP fragment was found to be highly expressed in human cells and was shown to synergize with the drug sorafenib to inhibit HCC cell growth and migration while promoting apoptosis.

Summary: Hepatoid adenocarcinoma of the lung (HAL) is a rare malignant tumor with characteristics resembling hepatocellular carcinoma but originating from the lung. Current major treatments for HAL include complete surgical resection and adjuvant chemotherapy, with some case reports suggesting potential benefits from immunotherapy and targeted therapy. Due to its rarity, there is a lack of systematic and extensive statistical research on HAL.
