Systems analysis of human responses to an aluminium hydroxide- adsorbed TLR7 agonist (AS37) adjuvanted vaccine reveals a dose- dependent and specific activation of the interferon-mediated antiviral response

Emilio Siena et al.

Summary: The candidate Adjuvant System AS37 contains a synthetic toll-like receptor agonist (TLR7a) adsorbed to alum, which promotes an immune response. In a study, healthy adults vaccinated with AS37-adjuvanted MenC-CRM197 conjugate vaccine showed early gene expression responses, changes in cytokine levels, and increased frequency of specific immune cells. The optimal dose for AS37 was found to be TLR7a 25-50 μg.

Polysaccharides derived from Chinese medicinal herbs: A promising choice of vaccine adjuvants

Xinhuan Wan et al.

Summary: The use of adjuvants in vaccines has been beneficial in promoting immune response and reducing production costs. Recently, polysaccharide adjuvants from Traditional Chinese medicine have gained attention for their immunomodulation, biocompatibility, and low toxicity properties. More research is needed to fully understand their mechanisms of action for clinical vaccine preparation.

Efficacy, safety, and immunogenicity of an inactivated, adjuvanted enterovirus 71 vaccine in infants and children: a multiregion, double-blind, randomised, placebo- controlled, phase 3 trial

Trong Toan Nguyen et al.

Summary: This study evaluated the efficacy, safety, and immunogenicity of the EV71vac vaccine in children aged 2-71 months. The results showed that the EV71vac vaccine had a 96.8% efficacy in preventing EV71-associated diseases and was well-tolerated. This study is of great importance for the health of children.

Enhanced Influenza Immunity by Nasal Mucosal Administration of the TPGS-Modified Liposomal Vaccine

Shijie Ma et al.

Summary: Liposomal influenza vaccines are a promising strategy to improve influenza protection. By incorporating cationic lipids and adding amphiphilic immune adjuvant TPGS, the vaccines can enhance cellular uptake and immune responses. Nasal mucosal immunization promotes a more robust immune response against influenza virus.

Type I and II interferons toward ideal vaccine and immunotherapy

Burcu Temizoz et al.

Summary: The immune system is equipped with interferons that play a crucial role in connecting innate and adaptive immunity to provide protective responses against pathogens and tumors. However, careful regulation of interferon production is necessary to prevent harmful effects on immune responses and the development of autoinflammatory or autoimmune conditions. Therapeutic strategies involving interferon-inducing adjuvants and immune-checkpoint blockers may hold promise for cancer treatment, while precise adjustment of timing, dose, and administration route is crucial for infectious and autoimmune diseases.
