Purification free N-glycan analysis by capillary zone electrophoresis: Hunt for the lost glycans

Summary: The possible sample losses during the purification process of fluorophore labeling reaction mixture in capillary gel electrophoresis were investigated. A normal polarity capillary zone electrophoresis separation mode and Trishexanoic acid running buffer at basic pH were applied for purification. The method effectively separated the labeled sample components of interest and showed electrophoretic profile differences caused by the cleanup process.

Labeling strategies for analysis of oligosaccharides and glycans by capillary electrophoresis

Summary: Capillary electrophoresis is widely used for oligosaccharide and glycan analysis, but the low electrophoretic mobility and lack of chromophore or fluorophore in saccharide molecules require modification before separation and detection. This review summarizes the labeling strategies used for oligosaccharide/glycan analysis, categorized based on the labeling mechanism and saccharide molecule. In addition to the commonly used 8-aminopyrene-1,3,6-trisulfonic acid trisodium salt, less common and recently developed tags applicable in glycomics are also summarized.

Advances in glycopeptide enrichment methods for the analysis of protein glycosylation over the past decade

Summary: Advancements in bioanalytical technology have sped up the analysis of complex protein glycosylation, benefiting the understanding of glycosylation in drug discovery and disease diagnosis. This review focuses on recent advances in glycosylation analysis, highlighting different strategies for glycopeptide enrichment and the challenges of O-glycosylation analysis. Improved enrichment methods combining different proteases for O-glycosylation analysis are also proposed.

Characterization of multi-cationic aminopyrene-based tag for oligosaccharide labeling by capillary electrophoresis with laser-induced fluorescence detection

Summary: In this study, a multi-cationic aminopyrene-based labeling tag was characterized for oligosaccharide analysis by capillary electrophoresis with laser-induced fluorescence detection. The APTMP labeling showed high reproducibility but lower reactivity compared to the commonly used APTS labeling. APTMP labeling remains promising for oligosaccharide labeling followed by CE-MS in a positive ion mode.
