O-GlcNAcylation modulates liquid-liquid phase separation of SynGAP/PSD-95

Summary: This study reveals that O-GlcNAcylation of SynGAP acts as a suppressor of LLPS of the SynGAP/PSD-95 complex. Through the identification of multiple O-GlcNAc modification sites and in vitro and cell-based assays, it is demonstrated that T1306 O-GlcNAc of SynGAP blocks the interaction with PSD-95, inhibiting LLPS. Additionally, O-GlcNAcylation suppresses LLPS in a dominant-negative manner, and O-GlcNAc cycling mediated by OGT and OGA regulates LLPS reversibly.
