Actin-driven chromosome clustering facilitates fast and complete chromosome capture in mammalian oocytes

Katarina Harasimov et al.

Summary: In meiosis, accurate chromosome segregation is crucial for reproduction. Human and porcine oocytes cluster their chromosomes temporarily before spindle assembly and chromosome segregation begin. The mechanism and function of chromosome clustering are still unknown. This study reveals that chromosome clustering is necessary to prevent chromosome losses during the long gap phase and to facilitate the rapid capture of all chromosomes by the acentrosomal spindle. The initial phase of clustering involves Formin-2- and Spire-nucleated actin cables, which interact with chromosomes and move them towards the center of the nucleus. Stable microtubule loops drive the late stages of chromosome clustering. Overall, this research highlights the critical role of chromosome clustering in accurate meiotic progression.

First pilot study of maternal spindle transfer for the treatment of repeated in vitro fertilization failures in couples with idiopathic infertility

Nuno Costa-Borges et al.

Summary: The study explored the technical feasibility of using maternal spindle transfer (MST) for the treatment of idiopathic infertility in patients with repeated in vitro fertilization (IVF) failures. MST was performed by transferring metaphase II spindles from patients' oocytes into donor oocytes, followed by IVF procedures. The study resulted in the birth of 6 apparently healthy children.

Summary: Forced mitophagy can reduce mtDNA carryover in mitochondrial replacement therapy by pre-labelling the mitochondrial outer membrane of a donor zygote with an autophagy receptor, as shown in mouse and human embryos.
