Autonomous rhythmic activity in glioma networks drives brain tumour growth

David Hausmann et al.

Summary: This study reveals the impact of network architecture and communication strategies on the biology of glioblastoma, a type of brain tumor. The findings demonstrate the crucial role of periodic Ca2+ activity in the network, and show that targeting this activity can significantly weaken global network communication and reduce tumor cell viability. The study highlights the vulnerability of glioblastoma networks and suggests the potential for developing novel therapies.

High-affinity anti-Arc nanobodies provide tools for structural and functional studies

Sigurbjorn Markusson et al.

Summary: Activity-regulated cytoskeleton-associated protein (Arc) plays a vital role in synaptic plasticity and memory formation. This study used high-affinity anti-Arc nanobodies to investigate the structure and function of Arc. The results suggest that the N-terminal domain of Arc is involved in membrane binding, while the C-terminal domain binds to postsynaptic protein ligands. Furthermore, the NTD and CTD are important for Arc oligomerisation and assembly of capsids.

Exosome-Mediated Immunosuppression in Tumor Microenvironments

Qi-Hui Xie et al.

Summary: Exosomes are membranous structures secreted by various cell types and serve as critical messengers for intercellular communication. In the tumor microenvironment, exosomes exert immunosuppressive effects, influencing tumor development and the effectiveness of immunotherapy. Therapeutic strategies targeting the molecular mechanism of exosome-mediated tumor development may overcome immune tolerance and contribute to innovative treatments.

Arc/Arg3.1 function in long-term synaptic plasticity: Emerging mechanisms and unresolved issues

Hongyu Zhang et al.

Summary: Arc is proposed as a critical regulator of long-term synaptic plasticity at excitatory synapses, with bidirectional control of synaptic strength by regulating AMPA-type glutamate receptor trafficking and actin cytoskeletal dynamics in dendritic spines. Key to this model, Arc is proposed to function as an activity-dependent regulator of AMPAR lateral membrane diffusion and trapping at synapses.

GAPDH controls extracellular vesicle biogenesis and enhances the therapeutic potential of EV mediated siRNA delivery to the brain

Ghulam Hassan Dar et al.

Summary: GAPDH, known as a housekeeping gene involved in glycolysis, also plays a crucial role in promoting vesicle clustering in endosomes and loading siRNA onto extracellular vesicles. This study demonstrates the significance of GAPDH in intercellular communication and drug delivery through EVs.

mRNA Trafficking in the Nervous System: A Key Mechanism of the Involvement of Activity-Regulated Cytoskeleton-Associated Protein (Arc) in Synaptic Plasticity

Michal Fila et al.

Summary: The protein Arc plays a crucial role in synaptic plasticity and memory consolidation by regulating synaptic activity. It may have evolved from the Ty3/Gypsy retrotransposon family, and is involved in synaptic activation through transcription induction and mRNA trafficking for localized translation.
