Peripheral glucocorticoid receptor antagonism by relacorilant with modest HPA axis disinhibition

Eva M. G. Viho et al.

Summary: Glucocorticoid stress hormones are essential for physiology and exert numerous actions via binding to the glucocorticoid receptor (GR). Relacorilant is a selective GR antagonist being evaluated for the treatment of endogenous Cushing's syndrome. This study compared the effects of relacorilant and the non-selective antagonist mifepristone, showing that relacorilant has potent antagonistic effects on GR signaling and prevents anti-inflammatory effects of glucocorticoids.

Parent Description of Anxiety in Angelman Syndrome

Christopher J. Keary et al.

Summary: Anxiety is increasingly being identified in individuals with Angelman syndrome (AS), with aggression being the most common behavioral manifestation of anxiety and separation from caregiver/parent being the most common circumstance. Individuals with a maternal deletion in AS had higher scores on anxiety, irritability, and hyperactivity scales, suggesting the need for adapting existing anxiety rating scales for use in AS. The Pediatric Anxiety Rating Scale was found to best capture behaviors described by caregivers in this study.

Cell type specificity of glucocorticoid signaling in the adult mouse hippocampus

Eva M. G. Viho et al.

Summary: Glucocorticoid stress hormones can affect mood and cognitive processes in the brain, with the hippocampus being a prominent target expressing GR and MR receptors. Analysis of single-cell RNA sequencing data confirmed cell-type specificity of GR and MR signaling, providing insights into cellular basis of glucocorticoid effects on hippocampal function.

Hippocampal glucocorticoid target genes associated with enhancement of memory consolidation

Jacobus C. Buurstede et al.

Summary: Glucocorticoids enhance memory consolidation of emotionally arousing events through genomic interactions in the hippocampus, affecting gene expression in both neuronal and non-neuronal cells. This effect is largely independent of training and involves differential DNA-binding of the glucocorticoid receptor and regulation of target genes.

A joint NCBI and EMBL-EBI transcript set for clinical genomics and research

Joannella Morales et al.

Summary: Comprehensive genome annotation is crucial for understanding clinically relevant variants, but the lack of standardized reporting and browser display complicates interpretation and reporting. To address this, Ensembl/GENCODE and RefSeq launched the MATCHED Annotation from NCBI and EMBL-EBI (MANE) collaboration to define universal standards for variant reporting and browser display. The MANE transcript sets provide representative transcripts for each human protein-coding gene, improving consistency and facilitating clinical interpretation.

Application of a pharmacological transcriptome filter identifies a shortlist of mouse glucocorticoid receptor target genes associated with memory consolidation

Jacobus C. Buurstede et al.

Summary: Glucocorticoids regulate memory consolidation by activating the glucocorticoid receptor and enhancing event-related memory. This study used pharmacological transcriptome filtering to identify eight genes associated with memory consolidation, providing insights into the mechanism of glucocorticoid effects.

Loss of nuclear UBE3A activity is the predominant cause of Angelman syndrome in individuals carrying UBE3A missense mutations

Stijn N. Bossuyt et al.

Summary: The research shows that mislocalization of UBE3A is the major cause of UBE3A dysfunction, followed by a loss of E3-ubiquitin ligase activity. Mutations affecting catalytic activity are found not only in the catalytic HECT domain, but also in the N-terminal half of UBE3A, suggesting an important contribution of this N-terminal region to its catalytic potential.
