Crosstalk between PI3K/AKT/mTOR and WNT/?-Catenin signaling in GBM-Could combination therapy checkmate the collusion?

S. Daisy Precilla et al.

Summary: This review focuses on the interplay between the PI3K/AKT/mTOR and WNT/beta-Catenin pathways in glioblastoma multiforme. It highlights the significance of these pathways in tumor proliferation, stemness, and chemotherapy resistance, and suggests the possibility of targeting them together to enhance the treatment of glioblastoma multiforme.

Cytokine chemokine network in tumor microenvironment: Impact on CSC properties and therapeutic applications

Mushtaq Ahmad Nengroo et al.

Summary: A subpopulation of cells, known as cancer stem cells (CSCs), in many cancers has stem cell traits and plays a role in metastasis and treatment resistance. These cells are regulated by interactions with components of the tumor microenvironment, such as mesenchymal stem cells, tumor related fibroblasts, adipocytes, endothelial cells, and immune cells, through a complex network of cytokines, chemokines, and growth factors. Inflammatory cytokines, including interleukin (IL)-1, IL-6, and IL-8, play a major role in these interactions. The activation of signaling pathways controlled by these cytokines increases the release of more cytokines, creating positive feedback loops that contribute to CSC self-renewal. Anti-inflammatory drugs have the potential to target CSCs by inhibiting signals from the tumor microenvironment and are considered as potential candidates for future cancer therapeutics.

Myeloid Cells in Glioblastoma Microenvironment

Alessandra De Leo et al.

Summary: Glioblastoma (GBM) is a highly aggressive and malignant brain tumor with a unique immune microenvironment that is resistant to immunotherapy. Myeloid cells play a crucial role in regulating immune and therapeutic responses in GBM. Studying the characteristics and functions of different populations of myeloid cells in GBM offers new insights for targeting specific cells for therapeutic purposes.

Imaging of Glioblastoma Tumor-Associated Myeloid Cells Using Nanobodies Targeting Signal Regulatory Protein Alpha

Karen De Vlaminck et al.

Summary: Single-cell RNA sequencing of human GBM samples revealed significant expression of SIRP alpha in myeloid cells, suggesting its potential as a target for nanobodies in mouse models. Analysis of different nanobody structures showed that small monovalent nanobodies have better tumor penetration and biodistribution characteristics.

Immunosuppression in Glioblastoma: Current Understanding and Therapeutic Implications

Benjamin T. Himes et al.

Summary: Glioblastoma (GBM) is the most common primary brain tumor in adults with a terrible prognosis. Immune suppression in many GBM patients has limited the effectiveness of immunotherapeutic agents, which have shown benefits in other cancers. Understanding tumor-mediated immune suppression is crucial for developing effective novel therapies for GBM, and overcoming this may be key for successful immunotherapy.

Key molecular alterations in endothelial cells in human glioblastoma uncovered through single-cell RNA sequencing

Yuan Xie et al.

Summary: By analyzing single-cell RNA sequencing data, researchers discovered different endothelial cell phenotypes in tumor vessels of human glioblastoma, which exhibit varying levels of activation and blood-brain barrier impairment. These findings are associated with different anatomical locations within and around the tumor.
