Genotype-phenotype associations in familial Mediterranean fever: a study of 500 Egyptian pediatric patients

Amal El Beshlawy et al.

Summary: This study investigates the genotype-phenotype associations of the three most common gene mutations in Egyptian FMF children. The results suggest that the genotype influences the clinical manifestations, disease severity, and response to colchicine.

Epigenetics and tissue immunity-Translating environmental cues into functional adaptations*

Zewen Kelvin Tuong et al.

Summary: Many immune cell subsets permanently reside in non-lymphoid organs, playing critical roles in tissue homeostasis and defense. Cell interactions and environmental cues shape immune cell function, and dysregulation can lead to inadequate immune responses. Understanding these mechanisms is crucial for studying tissue immunity.

Molecular mechanisms of transgenerational epigenetic inheritance

Maximilian H. Fitz-James et al.

Summary: Increasing evidence suggests that non-DNA sequence-based epigenetic information can be inherited across generations in various organisms. Recent research has uncovered the signals and mechanisms underlying these epimutations, which vary among species and have a limited effect in mammals. However, common principles are emerging, with transmission occurring through direct replication or indirect reconstruction of the signals. This research improves our understanding of the contribution of DNA sequence and epigenetic variation to heritable phenotypic differences.

Structural Insights into Methylated DNA Recognition by the Methyl-CpG Binding Domain of MBD6 from Arabidopsis thaliana

Yutaka Mahana et al.

Summary: This study determined the structure of the MBD domain of the MBD6 protein in Arabidopsis thaliana and investigated its binding properties to methylated DNA. The binding affinity of the MBD domain for methyl-CpG-containing DNA was found to be lower than that of mammalian MBD domains, due to the absence of positively charged residues. This research provides insights into how MBD proteins mediate epigenetic signals in plant cells.

Ethical implications of epigenetics in the era of personalized medicine

Josep Santalo et al.

Summary: This review discusses the ethical, legal, and social issues surrounding epigenetics research, with a focus on personalized medicine. It covers topics such as patient autonomy, data protection, social discrimination, and the distribution of health responsibilities. The review also highlights the technical and ethical challenges in epigenetic research and emphasizes the importance of integrating social experiences. It concludes with a reflection on the need to implement clinical epigenetics in society.

Evidence that direct inhibition of transcription factor binding is the prevailing mode of gene and repeat repression by DNA methylation

Sebastian Kaluscha et al.

Summary: Cytosine methylation efficiently silences CpG-rich regulatory regions of genes and repeats in mammalian genomes by directly inhibiting the binding of transcription factors rather than indirectly inhibiting via recruitment of methyl-CpG-binding domain proteins.

Transcriptional licensing is required for Pyrin inflammasome activation in human macrophages and bypassed by mutations causing familial Mediterranean fever

Matthew S. J. Mangan et al.

Summary: This study reveals that human monocyte-derived macrophages (hMDM) have a different regulation of Pyrin activation compared to monocytes and murine macrophages. Unlike the latter, hMDM fail to activate Pyrin in response to known Pyrin activators and require prolonged priming and increased Pyrin expression for Pyrin inflammasome response. However, FMF mutations lift the requirement for prolonged priming, enabling Pyrin activation without additional inflammatory signals. Additionally, the study unexpectedly finds that TcdB can activate the NLRP3 inflammasome in hMDM even without a Pyrin response.

Epigenetic Regulation of Inflammatory Signaling and Inflammation-Induced Cancer

Shawn Ying Xuan Tan et al.

Summary: This review discusses the interplay between epigenetics and inflammation. External and internal microenvironments contribute to epigenetic processes, and dysregulation of these processes can lead to cancer. Chronic inflammation plays a crucial role in the tumor microenvironment, and inflammatory signaling can modulate changes in the global epigenetic landscape.

Clinical and genetic characterization of familial Mediterranean fever among a cohort of Egyptian patients

Mohammed Hussien Ahmed et al.

Summary: This study aimed to evaluate FMF cases in Lower Egypt. The most commonly encountered mutation was the E148Q mutant allele, with a frequency of 31%. Patients with a positive family history and homozygous mutation had more attacks, greater severity, and higher amyloid deposition. Most patients required a dose between 1.5 and 3 mg/day.

Contribution of Dysregulated DNA Methylation to Autoimmunity

Samanta C. Funes et al.

Summary: Epigenetic mechanisms such as DNA methylation play a crucial role in autoimmune diseases like Rheumatoid Arthritis and Systemic Lupus Erythematosus, affecting disease activity through gene methylation. While the impact of epigenetic modifications on disease onset is recognized, further research is needed to establish the clear association between DNA methylation, gene expression, and disease pathogenesis.
