Regulation of redox signaling in HIF-1-dependent tumor angiogenesis

Valeria Manuelli et al.

Summary: This review provides an overview of the redox-sensitive steps in angiogenic signaling, the main molecular players involved, and their crosstalk with the unfolded protein response. New classes of inhibitors of these modulators which might act as antiangiogenic drugs in cancer are also discussed.

Regulation of glycolysis by the hypoxia-inducible factor (HIF): implications for cellular physiology

S. J. Kierans et al.

Summary: Under hypoxic conditions, eukaryotic cells can shift metabolism towards glycolysis to maintain ATP levels, a process regulated by HIF-1 alpha. This metabolic switch is crucial for cellular survival during acute hypoxic stress and has implications for cancer cell survival and growth. Understanding the mechanisms central to this reprogramming is important for both physiological and pathophysiological processes.

Hypoxia and hypoxia-inducible factors in diabetes and its complications

Sergiu-Bogdan Catrina et al.

Summary: HIFs regulate oxygen homeostasis, and insufficient activation of HIF signaling in diabetes impairs adaptive responses to hypoxia, due to inhibition of HIF-1 alpha stability and function by high glucose and elevated fatty acid levels. Targeting HIF signaling has potential for preventing and treating diabetes and related complications.

Cancer Cell Metabolism in Hypoxia: Role of HIF-1 as Key Regulator and Therapeutic Target

Summary: In cancer cells, metabolic reprogramming under low-oxygen conditions is crucial for tumor survival, with HIF-1 playing a pivotal role. Mitochondria are key players in regulating cellular energy and closely interact with HIF-1 to drive metabolic and functional changes in cancer cells.

Head and neck cancer: the role of anti-EGFR agents in the era of immunotherapy

Morena Fasano et al.

Summary: Head and neck cancers are the seventh most common cancer worldwide, with squamous cell carcinomas being the most frequent histologic subtype. The current standard treatment includes surgery or radiotherapy for early stage diseases, while locally advanced cases may require a more aggressive multi-modal approach. Anti-EGFR targeted therapy cetuximab is currently the only approved targeted therapy option, with the recent development of immune-checkpoint inhibitors providing new treatment options.
