Hyperglycemia, Reduced Hematopoietic Stem Cells, and Outcome of COVID-19

Benedetta Maria Bonora et al.

Summary: Admission hyperglycemia in COVID-19 patients is associated with reduced hematopoietic stem/progenitor cells (HSPCs), which predicts adverse outcomes. Mediation analysis reveals that the decreased HSPCs partially mediate the effect of hyperglycemia on COVID-19 outcome.

Bone marrow activation in response to metabolic syndrome and early atherosclerosis

Ana Devesa et al.

Summary: Experimental studies suggest that increased bone marrow activity is associated with cardiovascular risk factors, inflammation, and atherosclerosis. This study found that bone marrow activation is associated with metabolic syndrome and early atherosclerosis, even in the absence of systemic inflammation.

Impaired Hematopoietic Stem/Progenitor Cell Traffic and Multi-organ Damage in Diabetes

Gian Paolo Fadini et al.

Summary: During antenatal development, HSPCs arise and migrate to the fetal liver. In adulthood, HSPCs retain migratory activity and circulate to peripheral tissues. Diabetes mellitus jeopardizes HSPC traffic, reducing circulating HSPC levels and increasing the risk for multi-organ damage. The impaired traffic of HSPCs and enhanced generation of pro-inflammatory cells contribute to tissue damage and inflammation.

PATAS, a First-in-Class Therapeutic Peptide Biologic, Improves Whole-Body Insulin Resistance and Associated Comorbidities In Vivo

Edwige Schreyer et al.

Summary: Adipose tissue plays a crucial role in regulating whole-body metabolic fitness through controlling insulin sensitivity. Inactivation of ALMS1 leads to insulin resistance, and ALMS1 binds to PKC alpha, which is released upon insulin signaling. A peptide called PATAS promotes insulin-independent glucose absorption and de novo lipogenesis, and reduces insulin resistance and improves metabolic diseases.

Emily Bowers et al.

Summary: This review provides insights into how obesity leads to the accumulation of inflammatory cells, as well as the role of the HSC niche in hematopoiesis and inflammation, and its potential contribution to the hematopoietic response to obesity.

ALMS1 Regulates TGF-β Signaling and Morphology of Primary Cilia

Maria Alvarez-Satta et al.

Summary: This study reveals that ALMS1 depletion affects ciliary morphology and TGF-beta/BMP signaling, shedding light on its role in primary cilia function and cellular signaling, potentially linking it to the pathology of Alstrom syndrome.

Liver Fibrosis and Steatosis in Alstrom Syndrome: A Genetic Model for Metabolic Syndrome

Silvia Bettini et al.

Summary: Patients with Alström syndrome show significant abnormalities in liver fibrosis and steatosis, which may be related to metabolic and genetic alterations. Shear wave elastography and ultrasound sonographic are promising tools for evaluating disease progression. The ALMS1 gene may be involved in liver fibrogenesis.

Alstrom syndrome: an ultra-rare monogenic disorder as a model for insulin resistance, type 2 diabetes mellitus and obesity

Francesca Dassie et al.

Summary: Alstrom syndrome is a rare monogenic disorder caused by pathogenic variants of the ALMS1 gene, with common manifestations including retinal dystrophy, cardiomyopathy, obesity, deafness, insulin resistance, diabetes, fibrosis, and progressive renal or liver dysfunction. The ALMS1 protein is associated with the ciliary basal body and may play a role in ciliogenesis and normal ciliary function. Treatment options are currently limited to managing systemic diseases and lifestyle modifications for associated conditions like diabetes and obesity.
