Ubiquitylation of cyclin C by HACE1 regulates cisplatin-associated sensitivity in gastric cancer

Hong-Yue Jiang et al.

Summary: Cyclin C plays a crucial role in the regulation of cisplatin resistance in gastric cancer. The ubiquitylation of cyclin C by HACE1 facilitates mitochondrial fission and ROS production, leading to cell apoptosis. This finding provides new insight into the understanding of cisplatin resistance and its potential therapeutic targets in gastric cancer.

The giant E3 ligase HUWE1 is linked to tumorigenesis, spermatogenesis, intellectual disability, and inflammatory diseases

Lu Qi et al.

Summary: E3 ubiquitin ligase HUWE1 plays crucial roles in various physiological processes and its expanding substrate atlas poses challenges for potential therapeutic applications. Despite contradictory roles in tumor progression, HUWE1 also plays important roles in inflammasome activation and immune responses. This review summarizes the known substrates and functions of HUWE1 in different cell types and models, and discusses the importance of structural analysis in understanding its regulatory mechanisms.

Global cancer statistics 2020: GLOBOCAN estimates of incidence and mortality worldwide for 36 cancers in 185 countries

Hyuna Sung et al.

Summary: The global cancer burden in 2020 saw an estimated 19.3 million new cancer cases and almost 10.0 million cancer deaths. Female breast cancer surpassed lung cancer as the most commonly diagnosed cancer, while lung cancer remained the leading cause of cancer death. These trends are expected to rise in 2040, with transitioning countries experiencing a larger increase compared to transitioned countries due to demographic changes and risk factors associated with globalization and a growing economy. Efforts to improve cancer prevention measures and provision of cancer care in transitioning countries will be crucial for global cancer control.

Exploring the Other subfamily of HECT E3-ligases for therapeutic intervention

Sunil Singh et al.

Summary: The HECT E3 ligase family consists of three sub-families: NEDD4, HERC, and Other, with the latter being less explored. Members of the Other subfamily have diverse N-terminal regions preceding the conserved HECT domains, which influence the stability and activity of the proteins. They assemble atypical ubiquitin linkages and cooperate with E3 ligases from other subfamilies to form branched ubiquitin chains on substrates, making them potential candidates for therapeutic drug development.

Ubiquitin ligases in cancer: Functions and clinical potentials

Shanshan Duan et al.

Summary: Ubiquitylation is a crucial post-translational modification that regulates many cellular pathways. Dysregulated ubiquitin ligases play critical roles in cancer initiation and progression, acting as either tumor promoters or tumor suppressors depending on substrates and contexts. Current research focuses on understanding the regulatory roles of ubiquitin ligases and developing strategies for cancer therapy.
