Summary: Children with perinatal HIV have elevated levels of PD-1(+) CD8 T cells, which are associated with disease progression and coexpress other immune checkpoints. Initiation of antiretroviral therapy reduces total PD-1 levels and coexpression of multiple immune checkpoints. A subset of PD-1(+)2B4(+)CD160(+)TIM3(-) cells within PD-1(+) CD8 T cells predicts weaker HIV-specific proliferative responses, indicating functional exhaustion.

Markers of T Cell Exhaustion and Senescence and Their Relationship to Plasma TGF-β Levels in Treated HIV plus Immune Non-responders

Summary: HIV-infected immune non-responders (INR) exhibit higher proportions of CD4 T cell subsets expressing markers of exhaustion and senescence, as well as elevated plasma levels of IL-6, IP10, and sCD14. INR have higher levels of IP10 and lower levels of TGF-beta, with decreased cycling proportions of CD4 T regulatory cells. Proportions of T cells expressing TIGIT, PD-1, and CD57 positively correlate with IL-6 levels, while TGF-beta levels negatively correlate with proportions of TIGIT+ and PD-1+ T cell subsets.

Association between circulating exhausted CD4+ T cells with poor meningococcal C conjugate vaccine antibody response in HIV-infected children and adolescents

Summary: The study found that the co-expression of exhaustion markers and loss of co-stimulatory molecules influence the serum bactericidal antibody (SBA) response in HIV-infected patients. The frequency of exhausted CD4+ T cells was negatively correlated with SBA response, while IL-4 levels were positively correlated with SBA titers. Additionally, CXCL-13 levels were positively correlated with exhausted CD4+ T cells.
