Phosphorylation of NANOG by casein kinase I regulates embryonic stem cell self-renewal

Nicholas P. Mullin et al.

Summary: The self-renewal efficiency of mouse embryonic stem cells is controlled by the concentration of the transcription factor NANOG, which is phosphorylated by casein kinase I at specific residues. Phosphomimetic substitutions at certain phosphorylation sites within NANOG have site-specific functional effects, influencing DNA binding and LIF-independent self-renewal. Modeling the DNA-homeodomain complex helps explain the diverse effects of these substitutions, showing how phosphorylation may impact NANOG interactions crucial for ESC self-renewal.

OCT4 expression in human embryonic stem cells: spatio-temporal dynamics and fate transitions

L. E. Wadkin et al.

Summary: Enhancing the regulation of human embryonic stem cell pluripotency and differentiation trajectories is vital for their clinical applications. Analyzing the temporal and spatial dynamics of the pluripotency transcription factor OCT4 in hESC colonies reveals insights into its self-regulation and fluctuations within cells. Understanding the similarities in OCT4 expression among neighboring cells can provide valuable information about the behavior of proliferating hESC colonies.

CD29 Enriches for Cytotoxic Human CD4+ T Cells

Benoit P. Nicolet et al.

Summary: This study demonstrates that CD29 can be used to enrich for cytotoxic human CD4(+) T cells, which display a cytotoxic gene expression profile similar to CD8(+) T cells and produce proinflammatory cytokines and cytotoxic molecules. The expression of CD29 on CD4(+) T cells can also lead to target cell killing in vitro when transduced with a MART1-specific TCR.
