Extension of hydrogen borrowing alkylation reactions for the total synthesis of (-)-γ-lycorane

Summary: The total synthesis of (-)-gamma-lycorane (10 steps) and synthesis of (+/-)-gamma-lycorane (8 steps) from cyclohexenone was accomplished. A novel two-step hydrogen borrowing alkylation of an aziridinyl alcohol, coupled with a Ph* (Me5C6) deprotection/cyclisation procedure, was developed for the de novo formation of the fused 6,5 heterocyclic ring. This work represents one of the initial uses of hydrogen borrowing C-C bond formation as a key step in a total synthesis project.

A General Strategy for the Construction of Calyciphylline A-Type Alkaloids: Divergent Total Syntheses of (-)-Daphenylline and (-)-Himalensine A

Summary: An efficient general strategy for the synthesis of Daphniphyllum alkaloids has been established, leading to the total synthesis of (-)-daphenylline and (-)-himalensine A in 16 and 19 steps, respectively. The synthesis involves various reactions including an enantioselective Mg(ClO4)(2)-catalyzed intramolecular amidocyclization to construct the core structure, a Cu-catalyzed intramolecular cyclopropanation and subsequent phosphine-catalyzed Cope-type rearrangement for the himalensine A scaffold, and a one-pot Diels-Alder/aromatization method for the aromatic skeleton of daphenylline.

Additionally, a series of aromatic heterocyclefused piperidines were also obtained with excellent enantiocontrol by this methodology.
