Behavioural immune landscapes of inflammation

Summary: Transcriptional and proteomic profiling of individual cells have provided cellular landscapes of tissues, but fail to capture dynamic scenarios. This study used live imaging to record morpho-kinetic parameters of leukocytes in active inflammation and built behavioural landscapes that identified leukocyte identities and revealed a continuum of neutrophil states. Mutant screening identified the kinase Fgr as a driver of pathogenic inflammation, and interfering with Fgr protected mice from inflammatory injury.

Difference of immune cell infiltration between stable and unstable carotid artery atherosclerosis

Summary: This study compared the abundance and difference of specific immune cell subgroups in unstable atherosclerotic tissues, revealing a dominance of M2 macrophages, M0 macrophages, CD4 memory T cells, and CD8 T cells in the immune cell landscape of atherosclerosis. Unstable plaques showed a higher percentage of M2 macrophages compared to stable plaques. Additionally, correlation analysis suggested strong associations between certain immune cell types, such as naive CD4 T cells with gamma delta T cells and memory B cells with plasma cells.

Summary: Using WGCNA, significant modules related to SLE and PAH were identified, with shared genes showing a highly activated type I IFN response as a common feature in the pathophysiology of both diseases. Validation in another cohort confirmed these findings. A disease road model was proposed based on shared and unique gene signatures in SLE and PAH, suggesting a potential role for hsa-miR-146a in regulating IFN-induced genes and its importance in PAH secondary to SLE. This work highlights the high IFN response in SLE patients as a crucial susceptible factor for PAH and identifies novel gene candidates that could serve as biomarkers or therapeutic targets.
