Activation of p62-NRF2 Axis Protects against Doxorubicin-Induced Ferroptosis in Cardiomyocytes: A Novel Role and Molecular Mechanism of Resveratrol

Wei Yu et al.

Summary: Doxorubicin, a common chemotherapeutic agent, has limited clinical efficacy due to its severe cardiotoxicity. Resveratrol, known for its antioxidative and anti-inflammatory properties, protects cardiomyocytes by inhibiting ferroptosis induced by doxorubicin. This study reveals that resveratrol activates the p62-NRF2/HO-1 pathway and suppresses mitochondrial reactive oxygen species, thereby preventing ferroptosis in cardiomyocytes.

MiR-93-5p promotes granulosa cell apoptosis and ferroptosis by the NF-kB signaling pathway in polycystic ovary syndrome

Wei Tan et al.

Summary: This study found that miR-93-5p is upregulated in the granulosa cells of PCOS patients and promotes apoptosis and ferroptosis in these cells by negatively regulating the NF-kappa B signaling pathway. Silencing miR-93-5p can protect granulosa cell function, which is of great significance for improving the function of granulosa cells in PCOS patients.

Application of glutathione depletion in cancer therapy: Enhanced ROS-based therapy, ferroptosis, and chemotherapy

Boyi Niu et al.

Summary: Glutathione (GSH) plays a crucial role in cellular antioxidative system, particularly in cancer cells, where a high level of GSH is essential for scavenging ROS and detoxifying xenobiotics. Depletion of GSH has been shown to enhance the efficacy of various therapies such as ROS-based therapy, ferroptosis, and chemotherapy. This review comprehensively discusses the importance of GSH depletion in cancer therapy and various strategies employed to achieve it.

LPS Preconditioning Attenuates Apoptosis Mechanism by Inhibiting NF-κB and Caspase-3 Activity: TLR4 Pre-activation in the Signaling Pathway of LPS-Induced Neuroprotection

Pushpa Gandi Sangaran et al.

Summary: Neuroinflammation plays a key role in the progression of various neurodegenerative diseases, and recent studies have shown that LPS preconditioning provides neuroprotection by activating TLR4. Pre-exposure to low dose of LPS can confer a protective state against cellular apoptosis, indicating a role for TLR4 pre-activation in the signaling pathway of LPS-induced neuroprotection.

Crystal structures of the selenoprotein glutathione peroxidase 4 in its apo form and in complex with the covalently bound inhibitor ML162

Dieter Moosmayer et al.

Summary: Wild-type human glutathione peroxidase 4 (GPX4) was efficiently produced in human HEK cells with the co-expression of SBP2, leading to the determination of its crystal structure at 1.0 angstrom resolution. A mass-spectrometry-based approach was developed to monitor the reaction of the active-site selenocysteine with covalent inhibitors, enabling the determination of the crystal structure of GPX4 in complex with the covalent inhibitor ML162 [(S)-enantiomer]. This approach opens the possibility for further crystal structures of GPX4 in complex with potential cancer drug targets.

Glutathione peroxidase 4-regulated neutrophil ferroptosis induces systemic autoimmunity

Pengchong Li et al.

Summary: This study identifies a role for GPX4-regulated ferroptosis in neutrophils from SLE patients, triggered by interferons and autoreactive antibodies, contributing to lupus pathogenesis. The mechanism involves enhanced binding of a transcriptional repressor to the Gpx4 promoter leading to elevated lipid-reactive oxygen species. Neutrophil ferroptosis is shown to be a significant driver of neutropenia in SLE and heavily contributes to disease manifestations.
