Sustained release of tumor cell lysate and CpG from an injectable, cytotoxic hydrogel for melanoma immunotherapy

A platinum@polymer-catechol nanobraker enables radio-immunotherapy for crippling melanoma tumorigenesis, angiogenesis, and radioresistance

Summary: The interaction between PD-1 and PD-L1 in malignant melanoma cells promotes tumorigenesis, angiogenesis, and radio-resistance through mTOR hyperactivation. Blocking PD-L1 and mTOR signaling concurrently, along with radiotherapy, can alleviate tumor burden. A three-pronged platinum@polymer-catechol nanobraker, delivering mTOR inhibitor TAK228 and anti-PD-L1 antibody, effectively inhibits melanoma-PD-1-driven aggression and enhances melanoma eradication, while also amplifying the efficacy of radiotherapy.

Secretions from hypochlorous acid-treated tumor cells delivered in a melittin hydrogel potentiate cancer immunotherapy

Summary: This study demonstrates that different treatment methods can generate cell-derived secretions (CDS) with different effects. Among them, HOCl treatment generated CDS with the strongest stimulatory effect on immune cells. Loading HOCl-CDS into a hydrogel promoted tumor cell death and activation of immune cells, leading to inhibition of tumor growth and prolonged survival in mice. Furthermore, hydrogel-delivered HOCl-CDS enhanced the antitumor effects of immune checkpoint blockade.
