Rapid and sensitive SARS-CoV-2 detection using a homogeneous fluorescent immunosensor Quenchbody with crowding agents

Bo Zhu et al.

Summary: This study developed a homogeneous immunosensor based on a human anti-N protein antibody for SARS-CoV-2. The addition of a crowding agent significantly improved the response speed and sensitivity of the immunosensor. The developed immunosensor was validated in testing specimens from COVID-19-positive patients.

Brightening up Biology: Advances in Luciferase Systems for in Vivo Imaging

Shirley Liu et al.

Summary: Bioluminescence imaging (BLI) using luciferase reporters is a crucial method for visualizing cell populations and events in living animals, but its sensitivity remains limited for small numbers of cells in deep locations and larger animal models. Recent advances in luciferase systems aim to improve the sensitivity of in vivo BLI and expand its biological applications.

Fluorescence polarization immunoassay for the determination of diclofenac in wastewater

Anna Raysyan et al.

Summary: The study focused on the rapid quantification of DCF in wastewater through the synthesis of new tracer molecules and the evaluation of their performance. The sensitivity of the FPIA method for real wastewater samples was found to be high, with good agreement between the results and the reference method LC-MS/MS.

Facile and rapid detection of SARS-CoV-2 antibody based on a noncompetitive fluorescence polarization immunoassay in human serum samples

Keine Nishiyama et al.

Summary: This study presents an analytical method using fluorescence polarization immunoassay (FPIA) for rapid and quantitative detection of SARS-CoV-2 antibody in human serum. The FPIA system showed high accuracy in discriminating COVID-19 positive or negative serum samples. The total measurement time was about 20 minutes, and a serum volume of 0.25 μL was required, making it a rapid and convenient method for quantification of SARS-CoV-2 antibody.

One-step non-competitive fluorescence polarization immunoassay based on a Fab fragment for C-reactive protein quantification

Keine Nishiyama et al.

Summary: A non-competitive fluorescence polarization immunoassay (FPIA) using a Fab fragment was developed for large molecule quantification. The method, tested with C-reactive protein (CRP), achieved successful quantitative analysis without the need for sample pretreatment and with high sensitivity. The simplified procedure involved one-step mixing without the need for washing operations, enabling rapid protein quantification.

Evaluation and selection of potent fluorescent immunosensors by combining fluorescent peptide and nanobodies displayed on yeast surface

Akihito Inoue et al.

Summary: Quenchbody (Q-body) is a quench-based fluorescent immunosensor labeled with fluorescent dye(s) near the antigen-binding site of an antibody, capable of detecting a range of target molecules. Researchers developed a molecular display method of nanobody-based mini Q-bodies by combining yeast surface display and E4/K4 peptide-based fluorescence labeling. This approach allows for the quick selection of highly responsive mini Q-bodies and other fluorescent biosensors for various targets.

BRET Q-Body: A Ratiometric Quench-based Bioluminescent Immunosensor Made of Luciferase-Dye-Antibody Fusion with Enhanced Response

Riho Takahashi et al.

Summary: A quenchbody (Q-body) is an immunosensor that can recover fluorescence in the presence of an antigen, offering a simple and rapid system for antigen detection. The BRET Q-body, which fuses luciferase NanoLuc to a Q-body, can detect antigens based on the bioluminescence resonance energy transfer (BRET) principle and produce higher responses due to increased BRET efficiency. This new biosensor is promising for point-of-care tests without the need for an external light source.

Rapid construction of fluorescence quenching-based immunosensor Q-bodies using α-helical coiled-coil peptides

Takanobu Yasuda et al.

Summary: This study presents a rapid and efficient method for fabricating Quenchbodies (Q-bodies) using a stable coiled-coil peptide pair, allowing detection of targets with antigen-dependent fluorescence enhancement in various solutions. Additionally, FRET-type CQ-bodies using a dual-coloured peptide were described to enable more precise antigen quantification. The successful fabrication of nanobody-based CQ-body demonstrated its applicability to a range of antibody fragments.
