Comprehensive Profiling of an Aging Immune System Reveals Clonal GZMK+ CD8+ T Cells as Conserved Hallmark of Inflammaging

Denis A. Mogilenko et al.

Summary: This study characterized age-associated alterations in immune cells across multiple mouse organs and identified a distinct population of age-associated GZMK-expressing CD8(+) T cells. These Taa cells, highly clonal with specific features, were found to be a potential target to address age-associated dysfunctions of the immune system. In humans, proportions of the circulating GZMK(+) CDE3(+) T cell population that shares signatures with mouse Taa cells also increased during healthy aging.

TWIST1 induces phenotypic switching of vascular smooth muscle cells by downregulating p68 and microRNA-143/145

Jing Zhang et al.

Summary: TWIST1 protein plays a crucial role in the phenotypic switching of smooth muscle cells by decreasing microRNA-143/145 levels and repressing p68 expression. Research also indicates that TWIST1 triggers SMC phenotypic switching by promoting the degradation of p68 protein.

Esm1 and Stc1 as Angiogenic Factors Responsible for Protective Actions of Adipose-Derived Stem Cell Sheets on Chronic Heart Failure After Rat Myocardial Infarction

Mai Watanabe et al.

Summary: The study showed that transplantation of ADSC sheets improved cardiac function after myocardial infarction by reducing fibrosis and enhancing neovascularization. The transplanted ADSCs produced angiogenic factors such as Esm1 and Stc1, which may contribute to the improvement in cardiac function.

Somatic MAP3K3 and PIK3CA mutations in sporadic cerebral and spinal cord cavernous malformations

Tao Hong et al.

Summary: Cavernous malformations affecting the CNS are rare in the general population, with the majority being sporadic and associated with somatic missense variants in MAP3K3 and PIK3CA genes. The study identified distinct genetic subgroups with significant differences in gene expression and phenotype, with PIK3CA mutations potentially increasing the risk of hemorrhage.

Summary: This study successfully extracted cerebrovascular ECM from mouse and human brain tissues and identified over 1000 proteins in the ECM-enriched fraction. The identification of 147 core ECM proteins in the human brain vascular matrisome and the connection between brain ECM proteins and cerebrovascular diseases, provides unique mechanistic insight and potential drug targets for cerebrovascular diseases.

PIK3CA and CCM mutations fuel cavernomas through a cancer-like mechanism

Aileen A. Ren et al.

Summary: Vascular malformations are caused by increased signaling through the PI3K-mTOR pathway and loss of function of the CCM complex, which are found in the majority of human CCMs. Using mouse models, researchers discovered that the formation of CCMs can be effectively blocked by the mTORC1 inhibitor rapamycin.

Cerebral Cavernous Malformation From Mechanism to Therapy

Daniel A. Snellings et al.

Summary: Cerebral cavernous malformations are a common cause of central nervous system hemorrhage and stroke, and significant progress has been made in understanding the pathogenesis of this vascular disease over the past 20 years. New knowledge includes genetic and molecular causes of the disease, environmental influences on disease pathogenesis, and drivers of disease endpoints. These insights are the result of integrated research spanning human clinical studies, human genetic studies, studies in genetic models, and basic molecular and cellular studies.

De Novo Germline and Somatic Variants Convergently Promote Endothelial-to-Mesenchymal Transition in Simplex Brain Arteriovenous Malformation

Hao Li et al.

Summary: By analyzing a dataset from 269 patients, we found abnormal expression of endothelial and mesenchymal markers in bAVM lesions at both bulk and single-cell level. Additionally, de novo germline mutations affecting genes such as EXPH5 and vessel-related genes like EPAS1 and ENG were identified, with knockdown of epas1 in zebrafish embryos showing AVM-like phenotype exclusively in the brain. Furthermore, somatic KRAS mutations were detected in 72% of cases and shown to be associated with bleeding as the first symptom, with experimental studies demonstrating that KRAS mutations independently regulated EndMT features.

Structure and mechanism of blood-brain-barrier lipid transporter MFSD2A

Chase A. P. Wood et al.

Summary: MFSD2A is a crucial sodium-dependent lysophosphatidylcholine symporter responsible for importing docosahexaenoic acid into the brain, pivotal for brain development and function. Mutations in MFSD2A can lead to microcephaly syndromes. Understanding the structure and function of MFSD2A provides insight into its role as a transporter at the blood-brain barrier and potential for therapeutic modulation.

Somatic PIK3CA Mutations in Sporadic Cerebral Cavernous Malformations

Matthieu Peyre et al.

Summary: Researchers initially aimed to create a meningioma model but unexpectedly observed the development of cerebral cavernous malformations in engineered mice, leading them to test the implicated genes in patients with these malformations. Somatic mutations in PIK3CA were found to be more prevalent in tissue samples from sporadic CCMs compared to mutations in other genes, with a relatively small contribution from genes causing familial CCMs.

The Role of Mfsd2a in Nervous System Diseases

Bei Huang et al.

Summary: The major facilitator superfamily (MFS), particularly the member Mfsd2a, plays a critical role in maintaining blood-brain barrier integrity and transporting DHA in the study of nervous system diseases. While clinical research on Mfsd2a in nervous system diseases remains limited, experimental studies suggest promising effects of Mfsd2a in embryonic animals and cells.
