Differential Sphingosine-1-Phosphate Receptor-1 Protein Expression in the Dorsolateral Prefrontal Cortex Between Schizophrenia Type 1 and Type 2

Summary: Understanding and treating schizophrenia is challenging due to its heterogeneity. Recent research suggests that schizophrenia can be divided into two subtypes: Type 1 with relatively normal DLPFC transcriptome, and Type 2 with differentially expressed genes. S1PR1 is a gene that is highly upregulated in Type 2. By using receptor autoradiography and immunohistochemistry, it has been found that S1PR1 protein expression is significantly higher in Type 2 compared to Type 1 and controls. These findings suggest that PET S1PR1 can be used as a potential imaging biomarker to distinguish subtypes of schizophrenia.

Ponesimod, a second-generation S1P modulator, has shown high selectivity and rapid reversibility for S1P(1) receptor. It has been approved for the treatment of relapsing MS and offers advantages compared to other S1P receptor modulators.
