Emerging Strategies for Asymmetric Synthesis: Combining Enzyme Promiscuity and Photo-/Electro-redox Catalysis

Summary: Enzyme promiscuity has become an emerging opportunity in organic synthesis, as it allows for unnatural transformations of enzymes. The combination of enzyme catalysis and photo-/electro-redox catalysis offers numerous advantages, including new reactivity, high enantioselectivity, green synthesis, and high yields. Recent developments have demonstrated that combining enzyme promiscuity and photo-/electro-redox catalysis can unlock new asymmetric reactions. This mini-review highlights these emerging strategies and provides insights for the future development of this field.

It combines the bond formation capability of transition metal catalysis with the utility of photoinduced electron and energy transfer processes. Photocatalytic substrate activation allows the involvement of simple starting materials in metal-mediated bond-forming processes, while electron or energy transfer with organometallic intermediates provides a complementary activation mode to traditional catalytic platforms.

Chiral Photocatalyst Structures in Asymmetric Photochemical Synthesis

Merging the Non-Natural Catalytic Activity of Lipase and Electrosynthesis: Asymmetric Oxidative Cross-Coupling of Secondary Amines with Ketones

Summary: The enantioselective oxidative cross-coupling of secondary amines with ketones was achieved by combining the catalytic activity of lipase with electrosynthesis. This novel protocol allowed for the synthesis of 2,2-disubstituted 3-carbonyl indoles with a stereogenic quaternary carbon center in high yields and good enantio- and diastereoselectivities. The compatibility of hydrolase catalysis with electrosynthesis was demonstrated, highlighting the potential and mechanism of enzymatic electrosynthesis.

Stereo- and Enantioselective Benzylic C-H Alkenylation via Photoredox/Nickel Dual Catalysis

Summary: A stereo- and enantioselective benzylic C-H alkenylation via photoredox/nickel catalysis has been developed, allowing for smooth cross-coupling reactions of readily available alkylbenzenes and alkenyl bromides, even with complicated molecules. The reaction conditions are mild with good functional group tolerance, yielding chiral allylic compounds with high ee and E/Z ratio. Additionally, a triplet-energy-transfer-inhibiting strategy has been developed for stereoselective synthesis of alkenes under visible-light photocatalysis, with the coordination pattern of nickel with chiral bis-imidazoline ligand elucidated based on experiments and X-ray diffraction.
