Role of Azole Drugs in Promoting Fungal Cell Autophagy Revealed by an NIR Fluorescence-Based Theranostic Probe

Wanzhen Yang et al.

Summary: Autophagy is a widespread degradation system in eukaryotes, playing a crucial role in maintaining cellular homeostasis and substance recycling. In this study, a theranostic probe AF-1 with antifungal activity and autophagy-associated pH sensitivity was developed. AF-1 allowed visualization of the autophagy process in fungal cells and revealed the role of antifungal drugs in promoting autophagy to induce fungal cell apoptosis.

Bioorthogonal Coordination Polymer Nanoparticles with Aggregation-Induced Emission for Deep Tumor-Penetrating Radio- and Radiodynamic Therapy

Jingjing Liu et al.

Summary: Radiodynamic therapy (RDT) is a promising therapeutic approach for cancer treatment that combines ionizing irradiation and localized photodynamic therapy (PDT). Hf-AIE coordination polymer nanoparticles (CPNs) show strong RT and RDT effects under X-ray irradiation. Bioorthogonal click chemistry enhances tumor accumulation and retention of CPNs, leading to significant killing of cancer cells without noticeable biotoxicity.

Miae Won et al.

Summary: This study introduces a new construct, EA-BPS, which induces a synergistic antitumor effect through a combination of ROS production and GST-π inhibition, leading to enhanced cell-killing effect under hypoxic conditions. By making better use of available oxygen in tumor environments, improved therapeutic PDT outcomes can be achieved even in low oxygen conditions.

Aging Reprograms the Hematopoietic-Vascular Niche to Impede Regeneration and Promote Fibrosis

Yutian Chen et al.

Summary: Aging impairs regenerative capacity and leads to fibrosis in organs. Aberrant reprogramming of hematopoietic and vascular cell crosstalk in aging hinders regeneration and promotes fibrosis. Targeting aberrant endothelial node molecules may facilitate regeneration without scarring in the repair of multiple organs.

Cascade Reactions by Nitric Oxide and Hydrogen Radical for Anti-Hypoxia Photodynamic Therapy Using an Activatable Photosensitizer

Jian Sun et al.

Summary: In this study, a unimolecular glutathione and light coactivatable photosensitizer targeting mitochondria was developed, demonstrating high performance PDT effect through type I, type II reactions, and secondary radicals-participating reactions. With a relatively large two-photon absorption cross section for photoexcitation, the new photosensitizing system shows promising applications in both normoxic and hypoxic environments.

Cellular senescence in ageing: from mechanisms to therapeutic opportunities

Raffaella Di Micco et al.

Summary: Cellular senescence is characterized by permanent proliferation arrest and can occur in response to various stresses like telomere dysfunction. Apart from stress-induced senescence, it can also be part of controlled programs in biological processes like embryonic development. Senescent cells contribute to impaired tissue regeneration, chronic diseases, and organismal aging, but also present an opportunity for intervention in aging and associated diseases.

Senolysis by glutaminolysis inhibition ameliorates various age-associated disorders

Yoshikazu Johmura et al.

Summary: Research has shown that senescent cells rely on glutaminolysis for survival, and inhibiting this process can induce senolysis. Lysosomal membrane damage lowers intracellular pH in senescent cells, inducing KGA expression, leading to glutaminolysis, and ultimately producing ammonia to neutralize low pH and improve cell survival.

Two-Dimensional Design Strategy to Construct Smart Fluorescent Probes for the Precise Tracking of Senescence

Ying Gao et al.

Summary: The newly designed fluorescent probes for tracking cellular senescence and tissue aging have improved accuracy and discrimination by incorporating a second dimension of lysosomal pH. This unique design enables precise identification of SA-beta-gal and cancer-associated beta-gal, providing a new tool for studying cellular senescence. The crystal structures of a model enzyme complexed with each probe revealed the structural basis for probe recognition.
