Extraction Optimization and Antioxidant Activities of Ethanol Extract of the Brown Roll-Rim Mushroom Paxillus involutus (Agaricomycetes)

Box Behnken design (BBD) was used to optimize the ethanol extraction of Paxillus involutus (EPI). The optimal extraction conditions were found to be 45°C temperature, 1:35 solid:liquid ratio, and 5-hour extraction time, resulting in a yield of 13.57% EPI. In vitro evaluation of EPI's antioxidant activity showed comparable or close effects to the positive control vitamin C (VC) in scavenging DPPH, ABTS, and hydroxyl free radicals. In vivo evaluation on aging mice demonstrated that EPI had significant scavenging effects on malonic dialdehyde (MDA) and increased the activity of glutathione peroxidase (GSH-Px), superoxide dismutase (SOD), catalase (CAT), and total superoxide dismutase (T-SOD). Overall, EPI exhibited antioxidant effects in aging mice, indicating its potential as a natural antioxidant and in medical applications.

The antioxidant activity of EPI was next evaluated in vivo using aging mice; it was found to have appreciable effect on scavenging malonic dialdehyde (MDA) and could increase the ac-tivity of glutathione peroxidase (GSH-Px), superoxide dismutase (SOD), catalase (CAT), and total superoxide dismutase (T-SOD) in mice. Overall, EPI showed antioxidant effects in aging mice, thereby delaying aging, and has potential for application as a natural antioxidant and in medical applications.
