Along the road to crystal structure prediction (CSP) of pharmaceutical-like molecules

Summary: Computational methods for predicting crystal structures of organic compounds are mature and can be used routinely for rigid and semi-rigid molecules. Crystal structure prediction (CSP) is valuable in accelerating polymorph screening and reducing the risk of unexpected late-appearing polymorphs. However, it remains challenging when dealing with flexible molecules or high-Z' and multi-component crystal structures.

Analogy Powered by Prediction and Structural Invariants: Computationally Led Discovery of a Mesoporous Hydrogen-Bonded Organic Cage Crystal

Summary: This study used a combined approach to discover a rare low-density mesoporous hydrogen-bonded framework. The results show that this method can overcome the difficulties in comparing predicted molecular crystals with varying lattice parameters, providing a systematic approach for energy-structure comparison of chemically dissimilar molecules.

Ambiguous structure determination from powder data: four different structural models of 4,11-difluoroquinacriaone with similar X-ray powder patterns, fit to the PDF, SSNMR and DFT-D

Summary: Four different structural models, which all fit the same X-ray powder pattern, were obtained from unindexed X-ray powder data by a global fit. Despite their differences in lattice parameters, space groups, molecular packing, and hydrogen bond patterns, all models provided good Rietveld fits to the experimental data. Model B is likely to be the correct structure.

Development and assessment of an improved powder-diffraction-based method for molecular crystal structure similarity

Summary: Identifying whether two experimental crystal structures correspond to the same polymorph is a challenging problem in crystallography. In this study, a new PXRD-based similarity index and comparison method is proposed, which shows improved agreement compared to the popular COMPACK method. The strengths and weaknesses of each method are evaluated by analyzing the structure pairs for which they disagree. Using COMPACK and VC-PWDF in combination may be successful at narrowing the grey area of difficult-to-compare structures.

Digital navigation of energy-structure-function maps for hydrogen-bonded porous molecular crystals

Summary: Energy-structure-function (ESF) maps can facilitate the discovery of functional materials by predicting stable crystalline arrangements and functions of interest. The authors demonstrate a digital navigation protocol for ESF maps in the context of hydrogen-bonded organic frameworks constructed from triptycene- and spiro-biphenyl-based molecular building blocks.

Improved quantitative crystal-structure comparison using powder diffractograms via anisotropic volume correction

Summary: This study introduces a new method to improve crystal structure comparison by utilizing calculated PXRD, with an anisotropic volume correction technique to distinguish matching structures from other candidates. This method outperforms currently available PXRD comparison methods in separating similar structures and has demonstrated its effectiveness in distinguishing polytypes and identifying uncredited matching structures.
