Copper-Based Metal-Organic Framework Overcomes CancerChemoresistance through Systemically Disrupting DynamicallyBalanced Cellular Redox Homeostasis

Jia Liu et al.

Summary: This study presents a copper/catechol-based metal-organic framework that disrupts the redox homeostasis in drug-resistant cancer cells, leading to increased cellular oxidative stress and selective cytotoxicity. The framework effectively inhibits in vivo drug-resistant tumor growth and improves survival rates in tumor-bearing mice.

Synergizing effects of chemodynamic therapy and chemotherapy against breast cancer by oxaliplatin-loaded polydopamine/BSA@copper ferrite

Noor Alamer et al.

Summary: Chemodynamic therapy (CDT) involves the use of a Fenton reaction-based nanocatalyst to generate reactive oxygen species (ROS) inside tumor tissue. In this study, a copper ferrite nanoparticles (CuFe2O4) nanocatalyst was synthesized and combined with oxaliplatin (Oxa), a chemotherapeutic drug. The CuFe2O4.Oxa@PDA nanoparticles showed pH-responsive drug release and effectively inhibited breast cancer cell growth while exhibiting less cytotoxicity towards normal cells. The nanoparticles also catalyzed the Fenton reaction inside tumor cells, amplifying intracellular oxidative stress.

Red-Absorbing Ru(II) Polypyridyl Complexes with Biotin Targeting Spontaneously Assemble into Nanoparticles in Biological Media

Robin Vinck et al.

Summary: Four new ruthenium(II) polypyridyl complexes were synthesized and their properties were studied. It was found that these complexes could self-assemble into nanoparticles and accumulate in lysosomes upon internalization by cells. Conjugation with biotin increased cellular uptake but did not increase phototoxicity. However, these complexes showed high phototoxicity upon irradiation at long wavelengths (645-670 nm) and self-assembling behavior, making them promising for the development of new lysosome-targeted photosensitizers for photodynamic therapy.

Modified Aerotaxy for the Plug-in Manufacture of Cell-Penetrating Fenton Nanoagents for Reinforcing Chemodynamic Cancer Therapy

Kishwor Poudel et al.

Summary: The assembly of anisotropic nanomaterials has great potential in tumor therapeutics due to their ability to load active molecules and respond to external stimuli. Gas-phase epitaxy, also known as aerotaxy (AT), is an innovative method for the continuous assembly of anisotropic nanomaterials with uniform distribution. In this study, a modified AT method was developed to assemble cell-penetrating Fenton nanoagents, which showed enhanced cellular internalization capability.

Ferrocene-containing polymersome nanoreactors for synergistically amplified tumor-specific chemodynamic therapy

Yuheng Wang et al.

Summary: By loading GOD and TPZ in responsive polymersome nanoreactors, tumor acidic microenvironment can trigger the release of TPZ prodrugs, leading to the conversion of oxygen and glucose into H2O2 under the catalysis of ferrocene and subsequent generation of (OH)• radicals. This synergistic effect efficiently kills tumor cells and suppresses tumor growth, demonstrating the potential of ferrocene-containing responsive polymersome nanoreactors as a potent nano-platform for amplified CDT.
