A Multiple-Testing Procedure for High-Dimensional Mediation Hypotheses

Summary: Mediation analysis is gaining attention in epidemiologic studies and clinical trials, but existing methods have limitations. In this article, a multiple-testing procedure is proposed to accurately control error rates and improve power. Two data analysis examples demonstrate the effectiveness of the proposed method, and an R package is provided for implementation.

Large-Scale Hypothesis Testing for Causal Mediation Effects with Applications in Genome-wide Epigenetic Studies

Summary: The study addressed the issue of low power in popular statistical inference tests for causal mediation effects in genome-wide epigenetic studies, proposing a more powerful divide-aggregate composite-null test method that demonstrated improved power and Type I error rate control in finite samples. The new method was shown to outperform traditional tests like Sobel's test and the joint significance test for detecting mediation effects, providing more accurate results in identifying DNA methylation CpG sites mediating the effect of smoking on lung function.

A new approach to testing mediation of the microbiome at both the community and individual taxon levels

Summary: Motivation: Understanding the role of microbes in mediating the relationship between exposure and disease outcome is crucial for developing clinical interventions. However, existing methods have limitations in terms of controlling false discovery rate (FDR) and considering different types of null hypotheses. Results: We propose a new approach, LDM-med, based on inverse regression, which allows testing for mediation at both community and individual levels. Extensive simulations and application to a murine microbiome dataset demonstrate the effectiveness and flexibility of LDM-med.

Testing for Mediation Effect with Application to Human Microbiome Data
