Blocking iASPP/Nrf2/M-CSF axis improves anti-cancer effect of chemotherapy-induced senescence by attenuating M2 polarization

Hao Liu et al.

Summary: The complex interaction between cancer cells and the immune microenvironment plays a crucial role in tumor growth and treatment response. This study reveals a novel function of iASPP-Nrf2 in regulating the immune microenvironment during treatment-induced senescence. The accumulation of Nrf2 upon doxorubicin-induced senescence is mediated by increased cytoplasmic iASPP, which interrupts the Keap1/Nrf2 interaction and promotes Nrf2 stabilization and activation. Activated Nrf2 then transactivates a novel target gene, M-CSF, which induces macrophage polarization from M1 to M2 via a paracrine mechanism. Genetic inhibition of iASPP-Nrf2 suppresses the growth of apoptosis-resistant xenografts, highlighting the importance of M-CSF/M-CSFR-regulated macrophage polarization in these functional outcomes.

Summary: The transcription factor NRF2 plays a crucial role in regulating cytoprotection and metabolism. Its transient activation in healthy tissues prevents cancer initiation, but cancer cells hijack NRF2 to support their proliferation and develop resistance to treatment. Blocking NRF2 activity in tumors is a promising approach to counteract cancer progression and overcome treatment resistance.

Metabolic flexibility determines human NK cell functional fate in the tumor microenvironment

Sophie M. Poznanski et al.

Summary: NK cells play a central role in anti-tumor immunity, but their dysfunction in the tumor microenvironment is a major barrier for cancer immunotherapies. Research showed that human NK cell dysfunction in the tumor microenvironment is caused by suppression of glucose metabolism, and activating the antioxidant pathway can restore NK cell function and enhance anti-tumor activity. Additionally, expanded NK cells with metabolic flexibility not only sustained metabolic fitness but also improved tumor killing in response to nutrient deprivation, showing the potential for exploiting metabolic flexibility in cancer immunotherapy.

Heme catabolism by tumor-associated macrophages controls metastasis formation

Francesca Maria Consonni et al.

Summary: This study identifies a distinct subset of TAMs with high rates of heme catabolism by HO-1, playing a critical role in shaping a prometastatic tumor microenvironment and favoring immunosuppression, angiogenesis and epithelial-to-mesenchymal transition. These TAMs originate from BM precursors, accumulate in the blood of tumor bearers, and preferentially localize at the invasive margin through Nrf2 activation and NF-kappa B1-CSF1R-C3aR axis coordination. Hindering their recruitment or deletion of HO-1 inhibits metastasis formation and enhances anticancer immunotherapy, making them a potential antimetastatic target and prognostic blood marker.
