Autophagy promotes organelle clearance and organized cell separation of living root cap cells in Arabidopsis thaliana

Tatsuaki Goh et al.

Summary: The root cap is a tissue that guides root growth in plants. Its cells are constantly replaced through cell division and detachment. The transition of root cap cells from gravity-sensing cells to secretory cells involves rearrangement of organelles in the outer cell layer, which is partially regulated by autophagy. This developmentally regulated autophagy helps in the organized separation of living root cap cells, without causing immediate cell death.

Summary: This review discusses the role of phosphorylation in the regulation of autophagosome biogenesis and autophagic degradation in plants, and summarizes the experimentally verified phosphorylation sites in the core autophagy machinery in plants. Several approaches to test the roles of phosphorylation in the regulation of plant autophagy are also proposed.

V-ATPase is a universal regulator of LC3-associated phagocytosis and non-canonical autophagy

Kirsty M. Hooper et al.

Summary: This study reveals the important role of V-ATPase in non-canonical autophagy, as it is sufficient to activate CASM. V-ATPase recruits ATG16L1 during LAP, STING- and drug-induced CASM, indicating a common mechanism. Key molecular players, including NADPH oxidase/ROS, converge on V-ATPase during LAP. The study also demonstrates the contribution of CASM to the Salmonella host response and the disruption of V-ATPase-ATG16L1 axis by SopF.

Nondegradable ubiquitinated ATG9A organizes Golgi integrity and dynamics upon stresses

Qian Luo et al.

Summary: ATG9A is a membrane protein involved in the initiation of autophagy and plays a role in organizing Golgi dynamics along with GRASP55. Under heat stress, MARCH9 promotes the ubiquitination of ATG9A, leading to its dispersal from the Golgi apparatus to the cytoplasm and inhibiting GRASP55 oligomerization, resulting in Golgi fragmentation. Knockout of ATG9A or MARCH9 prevents Golgi fragmentation and protects Golgi functions under stress.

Selective autophagy regulates heat stress memory inArabidopsisby NBR1-mediated targeting of HSP90 and ROF1

Venkatesh P. Thirumalaikumar et al.

Summary: Plants experience various stresses in their growth process, and autophagy plays an important role in regulating plant recovery and memory of heat stress. NBR1 functions as a receptor for selective autophagy during recovery from heat stress, interacting with HSP90.1 and ROF1 to mediate protein degradation and modulate the response to heat stress. Loss of NBR1 function results in a stronger memory phenotype for heat stress in plants.

Beyond Autophagy: The Expanding Roles of ATG8 Proteins

Jose L. Nieto-Torres et al.

Summary: The ATG8 family proteins are key players in autophagy, participating in cargo recruitment, biogenesis, transport, and fusion with lysosomes. Recent studies have shown their involvement in diverse processes such as vesicle maturation and cargo specification, with complex regulatory mechanisms that are still not fully understood. Individual roles of different ATG8 family members in canonical or non-canonical pathways remain unclear. This review highlights the molecular questions surrounding ATG8s that are only beginning to be explored.

Global Warming, Climate Change, and Environmental Pollution: Recipe for a Multifactorial Stress Combination Disaster

Sara Zandalinas et al.

Summary: Studies have shown that an increase in different co-occurring multifactorial stress factors can result in a severe decline in plant growth and survival, as well as a decrease in microbiome biodiversity. This should serve as a warning for society to take action to reduce pollutants, combat global warming, and increase crop tolerance to multifactorial stress combinations.

Guidelines for the use and interpretation of assays for monitoring autophagy (4th edition)

Daniel J. Klionsky et al.

Summary: The guidelines presented in this article aim to assist researchers in selecting and interpreting methods to examine autophagy, as well as to help reviewers provide constructive critiques of reports focused on these processes. It emphasizes that no single assay is perfect for every situation, and multiple techniques may be needed to properly monitor autophagy in different experimental settings. The article also highlights the importance of targeting multiple autophagy-related genes in genetic approaches to block autophagy effectively.
