Prospective external validation of biomarkers to predict acute graft-versus-host disease severity

Marie Robin et al.

Summary: The study tested the ability of biomarker panels to predict outcomes for acute GVHD patients, showing slight improvement in predicting overall survival and nonrelapse mortality. The benefit of adding biomarkers to clinical parameters for nonresponse and mortality endpoints was found to be marginal.

Post-haematopoietic cell transplantation outcomes: why ST2 became a 'golden nugget' biomarker

Sophie Paczesny

Summary: Immunotherapies are promising for cancer treatment, but their clinical efficacy is limited by toxicities like GVHD. HCT is the most validated tumour immunotherapy, but its effectiveness is limited by GVHD and relapse. New cellular therapies and checkpoint inhibitors face similar challenges, but advancements in biomarker development post-HCT show promise in addressing these limitations.

MRD Tailored Therapy in AML: What We Have Learned So Far

Lok Lam Ngai et al.

Summary: Acute myeloid leukemia is a heterogeneous disease with poor survival, and many patient- and leukemia-specific characteristics have been shown to be valuable prognostic factors, with measurable residual disease (MRD) playing a crucial role in prognostic assessment. Multiparameter flow cytometry (MFC) based MRD measurement is currently the most commonly used method, with combinations of different techniques showing promising results in improving its prognostic value.

Elevation of Early Plasma Biomarkers in Patients with Clinical Risk Factors Predicts Increased Nonrelapse Mortality after Allogeneic Hematopoietic Stem Cell Transplantation

Masaaki Hotta et al.

Summary: Early prediction of nonrelapse mortality (NRM) in patients undergoing allogeneic hematopoietic stem cell transplantation (allo-HSCT) is challenging. Using plasma biomarkers ST2 and Reg3 alpha, along with the MAGIC algorithm, can help identify individuals at high risk for NRM. By analyzing blood samples and calculating risk scores, patients can be categorized into low-risk and high-risk groups for predicting the cumulative incidence of 6-month NRM.
