Summary: This study describes a practical chemical synthesis of E2-Ub-nucleosome conjugates that can trap the transient intermediates of E3-mediated Ub transfer from E2 to nucleosomes. The characterization and structure determination of these complexes reveal how different E3-E2 modules establish a specific architecture to orient the Ub towards the target lysine. This work provides the first example of using a chemical trapping strategy to study E3-mediated Ub transferring mechanisms on full-length and folded proteins.

Structural basis for activation of DNMT1

Chemical Synthesis of Post-Translationally Modified H2AX Reveals Redundancy in Interplay between Histone Phosphorylation, Ubiquitination, and Methylation on the Binding of 53BP1 with Nucleosomes

Summary: The chemical synthesis of homogeneously modified histones is a powerful approach to deciphering how post-translational modifications modulate epigenetic events. In this study, we developed a rapid method to synthesize phosphorylated and ubiquitinated H2AX proteins and found that H2AXS139 phosphorylation distorts the interaction interface between ubiquitin and the repair factor 53BP1. Our study revealed redundancy in the interplay of multiple histone PTMs, which is useful for controlling the dynamic distribution of effector proteins on nucleosomes.

Multistate structures of the MLL1-WRAD complex bound to H2B-ubiquitinated nucleosome

The study also uncovers different states of MLL1-WRAD during its assembly. The docking of subunits in the complex differs when bound to ubiquitinated nucleosomes, indicating the role of ubiquitin in promoting the formation of the active complex.
