Comprehensive Landscape of Immune Infiltration and Aberrant Pathway Activation in Ischemic Stroke

Rongrong Liu et al.

Summary: Ischemic stroke (IS) is a multifactorial disease caused by the interaction of multiple environmental and genetic risk factors. The immune microenvironment and inflammatory response play important roles in the occurrence and development of IS. The study found significant differences in the immune microenvironment between IS patients and controls, with increased inflammation-related pathway activity and decreased CD8+ T cell, B cell, and Th1 cell scores.

Pyroptosis in host defence against bacterial infection

Dominik Brokatzky et al.

Summary: Pyroptosis is a regulated form of cell death characterized by cell lysis and the release of pro-inflammatory molecules. It plays a crucial role in bacterial infection by promoting an inflammatory response and eliminating intracellular pathogens. Recent studies using various bacterial pathogens have enhanced our understanding of pyroptosis and its importance in host defense.

Robust In Vitro and In Vivo Immunosuppressive and Anti-inflammatory Properties of Inducible Caspase-9-mediated Apoptotic Mesenchymal Stromal/Stem Cell

Paola Alejandra Romecin et al.

Summary: Mesenchymal stromal stem/cells (MSCs) therapies have been widely used in the treatment of various disorders, but their mechanisms of action are not fully understood. Recent research has shown that infused MSCs undergo apoptosis induced by recipient cytotoxic T cells, which leads to immunomodulation. The use of MSC-derived extracellular vesicles (MSC-EVs) is also being explored as a cell-free therapeutic alternative. The introduction of the iCasp9/AP1903 suicide gene system into MSCs can induce apoptosis, potentially improving the safety of MSC-based therapies and overcoming current technical and biological uncertainties associated with MSC-EVs.

Mesenchymal stem cells attenuate liver fibrosis by targeting Ly6Chi/lo macrophages through activating the cytokine-paracrine and apoptotic pathways

Yuan-hui Li et al.

Summary: This study provides insights into the dual regulatory functions of bone marrow-derived MSCs (BM-MSCs) in attenuating hepatic fibrosis by promoting Ly6C(hi)/Ly6C(lo) subset conversion and Ly6C(lo) macrophage restoration through secreting antifibrogenic cytokines and activating the apoptotic pathway.

Efferocytosis induces macrophage proliferation to help resolve tissue injury

Brennan D. Gerlach et al.

Summary: This study demonstrates that nucleotides derived from apoptotic cell DNA hydrolysis by phagolysosomal DNase2a activate a pathway promoting non-inflammatory macrophage proliferation. The findings suggest that these nucleotides can amplify the resolution benefit by increasing the number of efferocytosing macrophages, thus enhancing tissue resolution in various experimental models including peritonitis, thymocyte apoptosis, and atherosclerosis regression.

Summary: Compared to psoriasis, patients with PsA have increased regulatory CD4+ T cells in peripheral blood and a specific population of CD8+CCR10+ T cells that are enriched in PsA. These CD8+CCR10+ T cells express a tissue-resident memory T cell transcriptional signature and a Tc2/22-like cytokine profile, suggesting a potential role in the development of PsA.

Summary: The study showed that DPSCs can regulate the inflammatory process by activating the PD-L1 pathway in response to inflammatory stimuli, reducing pro-inflammatory cytokines in pre-activated T lymphocytes. Their immunomodulatory/anti-inflammatory properties are closely related to their stemness status, highlighting the importance of communication with the inflammatory microenvironment.

New Insights on CD8+ T Cells in Inflammatory Bowel Disease and Therapeutic Approaches

Rosaely Casalegno Garduno et al.

Summary: Studies on the role of CD8(+) T cells in inflammatory bowel disease have shown contradictory outcomes, possibly due to the heterogeneity of the cells. Different subsets of CD8(+) T cells, such as Tc1 and Tc17 cells, may play a role in the pathogenesis of IBD. Regulatory CD8(+) T cells are abundant at sites of inflammation and can exhibit pro-inflammatory features.

Apoptotic cells represent a dynamic stem cell niche governing proliferation and tissue regeneration

Roi Ankawa et al.

Summary: The study shows that deletion of Caspase-9 in hair follicle stem cells attenuates the apoptotic cascade, resulting in significant temporal delays. These Casp9-deficient stem cells accumulate high levels of cleaved caspase-3 and are improperly cleared due to a caspase-3/caspase-9 feedforward loop. The apoptotic cells represent a dynamic stem cell niche that drives stem cell proliferation and tissue regeneration by continuously releasing Wnt3.
