Deep learning enables accurate analysis of images generated from droplet-based digital polymerase chain reaction (dPCR)

Summary: Accurate analysis of dPCR images is crucial for miniaturizing dPCR system for point-of-care testing. In this study, a deep learning model combined with circle Hough transform was used to perform semantic segmentation and count droplets in low-quality dPCR images. The modified deep learning model achieved rapid, absolute quantification and accurate identification of target DNA, similar to the commercial dPCR system. The deep learning-based approach also demonstrated high sensitivity for detecting pathogenic bacteria from low-quality images. These findings lay the groundwork for efficient nucleic acid quantification and establishment of a point-of-care analysis platform.

A Machine Learning and Computer Vision Approach to Rapidly Optimize Multiscale Droplet Generation

Two data-driven models, based on Bayesian regularised artificial neural network and XGBoost, accurately predict droplet size as a function of flow rates and surfactant properties. Compared to semi-empirical models, the data-driven models have higher accuracy. The models were also trained with simplified inputs, showing good prediction results. Additionally, a synthetic data set was generated, proving the feasibility of using this method in future lab on a chip applications.

It was concluded that the intervening HAMP domain passively transmits signals and shapes the signaling response depending on subtle changes in neighboring domains, leading to a richness in functional outputs as HKs vary in response to changing evolutionary pressures.
